线粒体动力学在消化系统肿瘤"炎-癌"转化方面作用的研究进展OA
Research advances on the role of mitochondrial dynamics in the"inflammation-cancer"transition of digestive system tumours
"炎-癌"转化是指慢性炎症微环境通过诱导基因组不稳定、抑制免疫监视、激活致癌信号通路等过程,最终导致细胞异常增殖,并发生恶性转化的生物学过程.线粒体是最基本的细胞器,其形态的动态改变会导致细胞内产生或长或短的片段,这种形态高度调节的过程被称为线粒体动力学.线粒体动力学调控着机体生理、病理状态下的细胞周期、新陈代谢、细胞存活与死亡等过程.线粒体动力学异常能放大炎症信号、加剧氧化应激、促进细胞损伤、诱导DNA突变,与消化系统肿瘤的"炎-癌"转化过程密切相关.本文从不同类型的消化系统肿瘤出发,系统总结线粒体动力学促进"炎-癌"转化的分子机制,旨在为基于线粒体动力学的靶向抗癌治疗提供实验依据和理论支持.
The"inflammation-cancer"transition denotes the biological process whereby chronic inflammatory microenviron-ments induce genomic instability,suppress immune surveillance,and activate carcinogenic signalling pathways,ultimately leading to abnormal cellular proliferation and malignant transformation.Mitochondria constitute fundamental organelles whose dynamic morphological alterations generate cellular fragments of varying lengths—a highly regulated process termed mitochondrial dynam-ics.Mitochondrial dynamics regulate cellular processes including the cell cycle,metabolism,survival,and death under both physi-ological and pathological conditions.Abnormal mitochondrial dynamics amplifies inflammatory signals,exacerbates oxidative stress,promotes cellular damage,and induces DNA mutations,being closely associated with the"inflammation-cancer"transfor-mation process in digestive system tumours.This paper systematically summarises the molecular mechanisms by which mitochon-drial dynamics facilitates"inflammation-cancer"conversion across different types of digestive system tumours.It aims to provide ex-perimental evidence and theoretical support for targeted anti-cancer therapies based on mitochondrial dynamics.
