lncRNA CASC2靶向miR-K11-3p调控内皮细胞的增殖、凋亡、迁移和侵袭OA
lncRNA CASC2 regulating the proliferation,apoptosis,migration,and invasion of endothelial cells through targeting miR-K11-3p
目的:公认的并广泛用于卡波西肉瘤研究的细胞株是人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs),研究长链非编码 RNA 癌易感性候选基因 2(long non-coding RNA cancer susceptibility candidate 2,lncRNA CASC2)靶向miR-K11-3p调控HUVECs增殖、凋亡、迁移和侵袭的分子机制.方法:qPCR检测lncRNA CASC2的表达水平;生物信息学预测lncRNA CASC2与miR-K11-3p的靶向关系并用双荧光素酶实验及qPCR加以验证;CCK-8实验检测细胞增殖水平;AnnexinV/Propidium Iodide实验检测细胞凋亡率;细胞划痕实验检测细胞迁移能力;Transwell 实验检测细胞侵袭能力.结果:pc DNA-CASC2组CASC2的表达水平明显高于Control组及pcDNA3.1组(P＜0.01);lncRNA CASC2靶向负调控miR-K11-3p的表达(P＜0.01);lncRNA CASC2抑制细胞增殖、促进细胞凋亡、降低细胞迁移及侵袭能力(P＜0.01);而外源回补 miR-K11-3p 可逆转 lncRNA CASC2 对内皮细胞的作用(P＜0.01).结论:lncRNA CASC2通过下调miR-K11-3p的表达,可抑制HUVECs的增殖、迁移和侵袭,并促进细胞凋亡.
Objective:The cell line recognized and widely used in Kaposi's sarcoma research is human umbilical vein endotheli-al cells(HUVECs),to explore the molecular mechanisms of long non-coding RNA cancer susceptibility candidate 2(lncRNA CASC2)on the proliferation,apoptosis,migration,and invasion of HUVECs by targeting miR-K11-3p.Methods:qPCR was used to detect the expression level of lncRNA CASC2;the targeting relationship between lncRNA CASC2 and miR-K11-3p was predicted by bioinformatics and verified by double-luciferase assay and qPCR;CCK-8 assay was used to detect cell proliferation;AnnexinV/Propidium Iodide assay was used to detect cell apoptosis rate;scratch assay was used to detect cell migration ability;Transwell assay was used to detect cell invasion ability.Results:The expression level of CASC2 in the pcDNA-CASC2 group was significantly higher than that in the Control group and pcDNA3.1 group(P＜0.01);lncRNA CASC2 negatively regulated the expression of miR-K11-3p(P＜0.01);lncRNA CASC2 inhibited cell proliferation,promoted cell apoptosis,and reduced cell mi-gration and invasion ability(P＜0.01);exogenously overexpressed miR-K11-3p reversed the effects of lncRNA CASC2 on endo-thelial cells(P＜0.01).Conclusion:LncRNA CASC2 can inhibit HUVECs'proliferation,migration and invasion,and promote cell apoptosis by down-regulating the expression of miR-K11-3p.
