为提高西蓝花中槲皮素、木犀草素和山奈酚的高效液相色谱-串联质谱(HPLC-MS/MS)检测效率,采用自制氮掺杂碳纤维布(NACC)与C18联合吸附净化基质中的杂质,优化了吸附剂类型与用量、净化温度、溶液pH,并对方法检测能力进行了评价.结果表明,该方法对三种目标物在50~5 000 μg·kg-1范围内具有良好的线性关系,决定系数R2为0.999 4~0.999 6;检出限分别为50、50、200 μg·kg-1,定量限分别为100、100、500 μg·kg-1;平均回收率为97.2%~99.2%,相对标准偏差为1.0%~1.3%.经单因素试验优化,确定C18用量为100 mg、NACC面积为2 cm2、净化温度为50℃、溶液pH为6时净化效果最佳,能够有效去除色素等干扰成分,同时保证目标物的高回收率与良好保留效果.实际样品检测中,该方法对目标物均表现出良好的检测效果和回收率.研究表明,该方法净化效率高、前处理简单、检测灵敏度高,可为植物性食品中黄酮类化合物的高效检测提供技术参考,同时拓展了 NACC材料在样品前处理中的应用潜力.

To improve the detection efficiency of quercetin,luteolin and kaempferol in broccoli by HPLC-MS/MS,self-prepared nitrogen-doped carbon cloth fiber(NACC)combined with C18 was adopted for adsorptive purification of matrix impurities.The type and dosage of adsorbents,purification temperature and solution pH were optimized,and the analytical performance of the method was evaluated.The results showed that the three target substances exhibited good linear relationships within the range of 50-5 000 μg·kg-1,with R2 ranging from 0.999 4 to 0.999 6.The limits of detection were 50,50 and 200 μg·kg-1,and the limits of quantification(LOQ)were 100,100 and 500 μg·kg-1,respectively.The average recoveries were 97.2%~99.2%,with relative standard deviation(RSDs)of 1.0%~1.3%.Single-factor experimental optimization confirmed the optimal purification conditions as follows:C18 dosage of 100 mg,NACC area of 2 cm2,purification temperature of 50 ℃,and solution pH of 6.Under these conditions,interfering components such as pigments could be effectively removed while ensuring high recoveries and satisfactory retention of target analytes.The proposed method presented favorable detection performance and recoveries in the analysis of practical broccoli samples.This study indicates that the established method features high purification efficiency,simple pretreatment and high detection sensitivity,which can provide a technical reference for the efficient determination of flavonoids in plant-based foods,and also expand the application potential of NACC materials in sample pre treatment.
