鸡白痢沙门菌噬菌体GSP040的生物学特性及基因组分析OA
Biological Characteristics and Genome Analysis of Salmonella Pullorum Phage GSP040
研究从鸡场粪便样本中分离纯化获得一株鸡白痢沙门菌烈性噬菌体,命名为GSP040.透射电镜观察结果显示,该噬菌体在形态上属于长尾噬菌体科,其病毒颗粒呈现出典型的二十面体头部和非收缩性尾部的形态特征.宿主谱测定表明,GSP040不仅可高效裂解鸡白痢沙门菌,对鼠伤寒沙门菌、肠炎沙门菌亦具有裂解活性.其最佳感染复数(multiplicity of infection,MOI)为0.1.一步生长曲线测定结果显示,其潜伏期为10 min,裂解期为80 min.该噬菌体具有良好的环境稳定性,在pH 3~11范围内及4~50 ℃条件下均能保持较高的裂解活性.全基因组测序结果表明,GSP040为双链DNA病毒,基因组全长为42 621 bp,GC含量为50%,编码69个预测的开放阅读框(open reading frames,ORFs),未检测到已知的毒力相关基因、耐药相关基因及溶原性相关基因.综上,GSP040是一株裂解谱广、裂解能力强、环境适应性优良的烈性噬菌体,在防控鸡白痢沙门菌等引起的沙门菌感染的生物制剂研发中,具有重要的研究价值与应用潜力.
A virulent phage against Salmonella Pullorum was isolated and purified from fecal samples of chicken farms and designated as GSP040.Transmission electron microscopy observation showed that the phage belonged to the Siphoviridae family in morphology,with virion characterized by a typical icosahedral head and a non-contractile tail.Host range analysis demonstrated that GSP040 could efficiently lyse Salmonella Pullorum,and also exhibited lytic activity against Salmonella Typhimurium and Salmonella Enteritidis.The optimal multiplicity of infection(MOI)was 0.1.The One-step growth curve analysis indicated that the latent period of 10 min and the burst period was 80 min.The phage possessed favorable environmental stability,maintaining high lytic activity at pH ranging from 3 to 11 and temperatures ranging from 4 ℃ to 50 ℃.Whole-genome sequencing results revealed that GSP040 was a double-stranded DNA virus with a full genome length of 42,621 bp and a GC content of 50%.A total of 69 predicted open reading frames(ORFs)were encoded,and no known virulence-related genes,drug resistance-related genes or lysogeny-related genes were detected.In conclusion,GSP040 is a virulent phage with a broad lytic spectrum,strong lytic capability,and excellent environmental adaptability,which has significant research value and application potential in the development of biological agents for the prevention and control of Salmonella infections caused by Salmonella Pullorum and other related strains.
