海伦兜兰组培苗生根培养体系优化OA
Optimization of Rooting Culture System for Paphiopedilum helenae Tissue Culture Seedlings
海伦兜兰(Paphiopedilum helenae)为国家Ⅰ级重点保护极小种群野生植物,其个体数量远低于最小存活种群临界值,面临随时灭绝的风险.为优化海伦兜兰组培苗生根培养体系,分析基本培养基、外源植物激素和有机添加物对组培苗生根的影响,筛选最适生根培养基.结果表明,基本培养基促进生根效果表现为1/2MS>MS>花宝1号;外源植物激素具有浓度效应,1.0 mg/L NAA和0.5 mg/L IBA均能有效促进生根,二者混合使用具有明显叠加效应;有机添加物以10%椰汁+5%香蕉为最佳组合,其次为15%椰汁和10%椰汁+0.2%蛋白胨,单独添加10%香蕉不利于生根.综合根系和植株生长状况,海伦兜兰组培苗最适生根培养基为1/2MS+1.8 mg/L 6-BA+1.0 mg/L NAA+0.5 mg/L IBA+10%椰汁+5%香蕉.
Paphiopedilum helenae is national first-class key protected wild plant with extremely small popula-tion.Number of its individuals is far below minimum viable population threshold,and it is at risk of extinction at any time.In order to optimize rooting culture system for P.helenae tissue culture seedlings,effects of basic media,exogenous plant hormones and organic additives on rooting of tissue culture seedlings were analyzed,and the most suitable rooting medium was selected.Results showed that rooting effect of basic media was in or-der of 1/2MS>MS>Hyponex No.1.Exogenous plant hormones had concentration effect,and both 1.0 mg/L NAA and 0.5 mg/L IBA could effectively promote rooting,combination of which had significant additive effect.The best combination of organic additives was 10%coconut water+5%banana,followed by 15%coconut water and 10%coconut water+0.2%peptone,and addition of 10%banana alone was not conducive to rooting.Synthesiz-ing growth conditions of roots and plants,the most suitable rooting medium for P.helenae tissue culture seed-lings was 1/2MS+1.8 mg/L 6-BA+1.0 mg/L NAA+0.5 mg/L IBA+10%coconut water+5%banana.
