井冈山杜鹃(Rhododendron jingangshanicum Tam)为高山特有观赏植物,具有较高的观赏和应用价值,因生境破坏和人为干扰面临生存威胁.为探究其对环境适应的分子基础,沿海拔梯度(1068-1787 m)采集样本进行转录组测序.组装获得 230768 条 Unigene,其中 55.23%(127455 条)在 NR 数据库获得注释,51605 条和 48401 条分别获得 GO 和 KEGG 注释.功能分析显示,植物-病原菌互作、MAPK 信号通路和氧化磷酸化等通路显著富集.差异表达分析共鉴定 34714 个 DEGs,其中 RJ1345 vs RJ1504 和 RJ1068 vs RJ1787 组均表现出高转录组变化,而 RJ1504 vs RJ1682 组 DEGs 显著较少(1385 个),提示基因表达对海拔的响应呈现非线性特征.转录因子家族共预测 2906 个转录因子,bHLH(9.87%)和 MYB-related(8.01%)家族成员最多.SSR 分析检测到67541 个位点,以A/T(43.59%)和AG/CT(34.31%)重复为主.研究结果丰富了井冈山杜鹃的基因信息,可为后续遗传多样性分析和遗传资源保护策略的制定提供参考.

Rhododendron jingangshanicum Tam is an alpine ornamental plant with high ornamental and application value.It faces sur-vival threats due to habitat destruction and human disturbance.In order to explore the molecular basis of environmental adaptation and thereby enhance the conservation and effective utilization of the germplasm resources.In this study,we performed transcriptome sequencing of R.jingangshanicum Tam samples along an elevational gradient(1068-1787 m).A total of 230768 Unigenes were as-sembled,of which 55.23%(127455)were annotated in the NR database,51605 and 48401 were annotated by GO and KEGG data-bases,respectively.Functional analysis showed that plant-pathogen interactions,MAPK signaling and oxidative phosphorylation were significantly enriched.A total of 34714 DEGs were identified by differential expression analysis.Among them,RJ1345 vs RJ1504 and RJ1068 vs RJ1787 showed high transcriptome variations,while RJ1504 vs RJ1682 showed significantly fewer DEGs(1385),sugges-ting that the response of gene expression to elevation showed non-linear characteristics.A total of 2906 transcription factors were pre-dicted in the transcription factor family,bHLH(9.87%)and MYB-related(8.01%)family members were the most.A total of 67541 SSR sites were detected by SSR locus analysis,mainly A/T(43.59%)and AG/CT(34.31%)repeats.The results of this study en-riched the genetic information of R.jingangshanicum Tam,and provided a theoretical basis for the subsequent research on the analy-sis of species genetic diversity and the formulation of effective genetic resource protection strategies.
