[目的]探明铅镉胁迫对不同羊肚菌的损伤规律,为羊肚菌耐重金属胁迫菌株选育提供依据.[方法]采用不同浓度铅、镉重金属培养基(2 mg/L、4 mg/L、6 mg/L、8 mg/L、10 mg/L)对山东 LM、乐丰 13、DB33-5等 9个羊肚菌菌株进行培养,研究铅和镉胁迫对不同羊肚菌菌丝生长速度、菌落形态、抑菌率、生物量、重金属含量的影响.[结果]随铅、镉浓度提高,所有羊肚菌菌丝生长速度整体呈下降趋势;铅浓度 10 mg/L时,山东 LM菌丝生长速度达 7.35 mm/d,显著高于其余菌株;镉浓度 10 mg/L时,山东 LM菌丝生长速度仍在 4.00 mm/d以 上,DB8-4其次,为 3.82 mm/d.菌落直径随铅浓度升高不断减小,菌丝密度降低,气生菌丝变少,部分菌株菌丝颜色变黄,乐丰 13在铅浓度 6~10 mg/L时形成 1个不规则抑菌圈;高浓度镉胁迫下部分羊肚菌菌落几乎停止生长.随铅、镉浓度升高,羊肚菌菌丝生物量整体呈下降趋势;镉胁迫下羊肚菌菌丝生物量普遍低于铅胁迫.随铅浓度增加所有羊肚菌铅含量均上升,DB29-2在铅浓度 10 mg/L时菌丝铅含量最高,达 3 991.67 mg/kg;随镉浓度增加,羊肚菌菌丝镉含量出现两类情况,一类是黄色支系羊肚菌菌丝镉含量不断上升;另一类是大部分黑色支系羊肚菌菌丝镉含量先升后降,DB41-1在镉浓度 10 mg/L时菌丝镉含量最高,达 11 194.02 mg/kg.[结论]高浓度镉对羊肚菌菌丝生长速度和生物量抑制作用较铅更强;羊肚菌镉富集能力高于铅;黄色支系羊肚菌DB8-4的菌丝生长速度耐受性较强;山东LM耐镉富集能力较强.

[Objective]The effect of Cd and Pb stress on damage of different Morchella strains was investigated to provide a scientific basis for breeding Morchella strains with a higher tolerance to heavy metals stress.[Method]9 Morchella strains of Shangdong LM,Lefeng 13,DB33-5,and so on were cultured on media containing Pb and Cd with five different concentrations of 2 mg/L,4 mg/L,6 mg/L,8 mg/L and 10 mg/L to investigate the effects of Pb and Cd stress on growth rate,colonial morphology,antibacterial rate,biomass and heavy metal content of mycelia of different Morchella strains.[Result]The mycelium growth rate of 9 Morchella strains showed a general downward trend with increase of Pb and Cd concentration.When Pb concentration was 10 mg/L,the mycelium growth rate of Shangdong LM strain reached 7.35 mm/d,significantly higher than other strains.The mycelium growth rate of Shangdong LM and DB8-4 strain still kept more than 4.00 mm/d and 3.82 mm/d when Cd concentration was 10 mg/L respectively.With increase of Pb concentration,the colonial density decreased continuously,its aerial mycelia reduced,and the mycelium color of some Morchella strains became yellow.The mycelium of Lefeng 13 Morchella strain formed an irregular inhibition zone when Pb concentration was 6-10 mg/L.The colony of some Morchella strains stopped growing almost under high Cd concentration.With increase of Pb and Cd concentration,the mycelial biomass of 9 Morchella strains showed a declining trend overall.The mycelial biomass of 9 Morchella strains under Cd stress was lower than that under Pb stress generally.The Pb content in mycelial of 9 Morchella strains rose with increase of Pb concentration,and the Pb content in mycelial of DB29-2 strain reached 3 991.67 mg/kg(the highest)when Pb concentration was 10 mg/L.The Cd content in mycelial of 9 Morchella strains showed two different patterns with increase of Cd concentration.The Cd content in mycelial of Morchella strains belonging to yellow clade and black clade rose continuously and showed an increasing first and then decreasing trend with increase of Cd concentration respectively.The Cd content in mycelial of DB41-1 strain reached 11 194.02 mg/kg when Cd concentration was 10 mg/L.[Conclusion]The high Cd concentration has the stronger inhibitory effects on mycelium growth rate and biomass of 9 Morchella strains compared with those of high Pb concentration.The Cd enrichment capacity of 9 Morchella strains is higher than their Pb enrichment capacity.DB8-4 Morchella strain belonging to yellow clade has a higher tolerance to mycelial growth rate.Shangdong LM Morchella strain has the higher Cd enrichment capacity among 9 Morchella strains.
