[目的]探明姬竹叶石斛(Dendrobium odiosum Finet)的叶绿体基因组特征,为其植物学分类、遗传进化研究及开发利用提供参考.[方法]结合高通量测序和三代 ONT测序技术获得姬竹叶石斛叶绿体基因组序列,分析其叶绿体基因组的结构、特征及系统进化关系.[结果]姬竹叶石斛叶绿体基因组具有典型的四分体结构,序列长度为 157 312 bp,共含有 116条 Unigenes,含 8个 rRNA、38个 tRNA和 70个编码基因.共检测出 63个 SSR位点,其中,以单核苷酸、二核苷酸为主,碱基组成多为 A、T重复单元.姬竹叶石斛cpDNA序列编码 18 061个氨基酸,CDS序列密码子偏好使用的密码子多以 A/U(T)碱基结尾.姬竹叶石斛与竹枝石斛(Dendrobium salaccense)、美花石斛(Dendrobium loddigesii)亲缘关系最近,与形态相似的细叶石斛(Dendrobium hancockii)亲缘关系较远,二者 IR/SC边界存在明显差异.[结论]姬竹叶石斛叶绿体基因组具有典型的四分体结构,与细叶石斛亲缘关系较远,推测姬竹叶石斛是不同于细叶石斛的单独物种.

[Objective]The chloroplast genome characteristics of Dendrobium odiosum was investigated to provide a reference for botanic classification,genetic evolution,development and utilization of D.odiosum.[Method]The chloroplast genome sequence of D.odiosum obtained by high-throughput sequencing and third generation ONT sequencing technology was used to analyze the structure,characteristics and phyletic evolution relationship of the chloroplast genome in D.odiosum.[Result]The D.odiosum chloroplast genome with a typical tetrad structure and 157 312 bp of sequence length contained 116 unigenes,8 rRNAs,38 tRNAs and 70 protein-coding genes.63 detected SSR sites were mainly mono-and di-nucleotides,and the base compositions were mostly A and T repetitive units.There were 18 061 amino acids in cpDNA coding sequence of D.odiosum.The codons of CDS sequence preferred to end with A/U(T)bases.There was a close genetic relationship between D.odiosum,Dendrobium salaccense and Dendrobium loddigesii.There was a distant genetic relationship between D.odiosum and Dendrobium hancockii and a significant difference in the IR/SC boundary between D.odiosum and D.hancockii.[Conclusion]The chloroplast genome of D.odiosum has a typical tetrad structure,and there is a distant genetic relationship between D.odiosum and D.hancockii.It is speculated that D.odiosum is a separate species.
