小分子泛素样修饰(Small molecule ubiquitin-like modification,SUMO)是一种动态可逆的蛋白质翻译后修饰,通过精确调控底物蛋白的活性、定位及互作,深度参与免疫与炎症反应的调节过程.梳理了 SUMO 化修饰及其抑制在免疫调控中的双面作用,一方面,SUMO 化可促进抑制性复合物的组装,限制炎症基因表达并推动巨噬细胞向M2 型抗炎表型极化,从而发挥抗炎与维持稳态的作用;另一方面,也可通过稳定NLRP3 炎症小体,促进Th17 分化等机制加剧炎症.反之,SUMO化抑制通常可解除对I型干扰素通路等天然免疫信号的负向调控,增强机体抗病毒与抗肿瘤免疫,但在特定情境下同样可能激活抗炎通路.对该修饰机制的解析,不仅深化了免疫动态调节网络的认识,也为感染、自身免疫病及肿瘤等疾病的靶向干预提供了分子靶点与潜在治疗策略,有助于推动基础免疫研究向临床应用的转化.

Small molecule ubiquitin-like modification(SUMO)is a dynamic and reversible post-translational modification that profoundly regulates immune and inflammatory responses by precisely modulating the activity,localization and interactions of substrate proteins.Elucidates the dual roles of SUMOylation modification and its inhibition in immune regulation.On one hand,SUMOylation can exert anti-inflammatory and homeostatic functions by promoting the assembly of inhibitory complexes,restricting the expression of inflammatory genes,and driving macrophage polarization towards the M2 anti-inflammatory phenotype.On the other hand,it can also exacerbate inflammation through mechanisms such as stabilizing the NLRP3 inflammasome and promoting Th17 cell differentiation.Conversely,the inhibition mediated by SUMOylation typically relieves the negative regulation of innate immune signaling pathways,such as the type I interferon pathway,thereby enhancing antiviral and antitumor immunity,however,in specific contexts,it may also activate anti-inflammatory pathways.The analysis of this regulatory mechanism not only advances understanding of the dynamic immune regulatory network but also provides molecular targets and potential therapeutic strategies for the targeted intervention of infectious diseases,autoimmune disorders and cancers,thereby facilitating the translation of basic immunological research into clinical applications.
