紫草素诱导肝癌细胞凋亡和自噬效应及其对ROS水平的影响OA
Shikonin Induces Apoptosis and Autophagy in Liver Cancer Cells and Its Ef-fect on ROS Levels
目的 探讨紫草素抑制肝癌细胞SMMC-7721的作用机制.方法 体外培养SMMC-7721细胞,不同浓度紫草素处理.MTT法检测72 h后细胞相对活力抑制率;台盼蓝拒染法评估24 h细胞存活率;流式细胞术分析不同浓度紫草素处理组的凋亡率、ROS水平、自噬活性及细胞周期分布.结果 紫草素以浓度依赖方式抑制细胞相对活力(IC₅₀为1.25 mg/L).与对照组相比,0.25 mg/L组早期凋亡率显著升高(26.89%vs.1.87%,P<0.05);1和4 mg/L组自噬荧光强度均值分别为12.63%和11.78%(对照组7.96%,P<0.05).4 mg/L组活细胞比例由95.59%下降至42.45%;ROS水平在1、4 mg/L组显著升高(P<0.05);4 mg/L紫草素使SubG₁期细胞比例上升52.22%,S期及G₂/M期比例下降(P<0.01).结论 紫草素可抑制肝癌细胞相对活力,并诱导凋亡及自噬发生,其效应可能与ROS水平升高相关.
Objective To explore the mechanism of action of purpurin in inhibiting liver cancer cell line SMMC-7721.Methods SMMC-7721 cells were cultured in vitro and treated with different concentrations of shikonin.The inhibitory rate of relative cell viability was as-sessed using the MTT assay after 72 hours;cell survival rate was evaluated via trypan blue exclusion assay after 24 hours;flow cytometry was employed to analyze apoptosis rate,ROS levels,autophagy activity,and cell cycle distribution in groups treated with different concen-trations of shikonin.Results Shikonin inhibited relative cell viability in a concentration-dependent manner(IC₅₀=1.25 mg/L).Com-pared with the control group,the early apoptosis rate significantly increased in the 0.25 mg/L group(26.89%vs.1.87%,P<0.05);the mean autophagy fluorescence intensities in the 1 and 4 concentration groups were 12.63%and 11.78%,respectively(control group:7.96%,P<0.05).The proportion of viable cells in the 4 concentration group decreased from 95.59%to 42.45%.ROS levels significantly increased in the 1 mg/L and 4 mg/L groups(P<0.05).Treatment with 4 mg/L shikonin increased the proportion of SubG ₁ phase cells(52.22%)and decreased the proportions of S phase and G ₂/M phase cells(P<0.01).Conclusion Shikonin can inhibit the relative vi-ability of hepatocellular carcinoma cells and induce apoptosis and autophagy,effects that may be related to increased ROS levels.
