生长年限对蒙古黄芪根际土壤环境的影响OA
Effects of Growth Years on the Rhizosphere Soil Environment of A.mongholicus
以蒙古黄芪 Astragalus membranaceus(Fisch.)Bge.var.Mongholicus(Bge.)Hsiao种子和种苗为试材,采用16S rRNA微生物多样性测序和LC-MS代谢组学等方法,研究了生长年限对蒙古黄芪根际土壤细菌群落多样性、代谢物和酶活性的影响,以期为蒙古黄芪的生态栽培提供参考依据.结果表明:1)Alpha多样性分析发现,随着生长年限的增加,蒙古黄芪根际土壤细菌群落Observed species指数和Chao1指数出现了先下降后升高的趋势,而Shannon指数和Simpson指数变化趋势不大;Beta多样性分析发现,在属水平下,H1-vs-HCK、H2-vs-HCK和H1-vs-H2中分别有47、26种和12种差异物种;其预测的功能基因分别富集到类胡萝卜素生物合成、甘油脂代谢和甘油磷脂代谢等KEGG通路.2)代谢组学分析发现,H1-vs-HCK、H2-vs-HCK和H1-vs-H2中分别有538、225种和559种差异代谢物,且分别富集到128、70条和150条KEGG通路;相关性分析发现部分差异物种的相对丰度与差异代谢物的表达量达到显著.3)酶活性分析发现,生长年限的增加降低了根际土壤的过氧化氢酶、碱性磷酸酶和脲酶的活性,提高了蔗糖酶活性;相关性分析发现,细菌群落的多样性与部分酶的活性有着高度的显著相关.综上所述,蒙古黄芪种子和种苗在生长发育期间对其根际土壤细菌群落多样性、代谢物和酶活性均产生了不同的影响.
Taking Astragalus membranaceus(Fisch.)Bge.var.Mongholicus(Bge.)Hsiao(A.mongholicus)seeds and seedlings as test materials,the effects of growth years on the bacterial community diversity,metabolites,and enzyme activity of A.mongholicus rhizosphere soil were studied by using 16S rRNA microbial diversity sequencing and LC-MS metabolomics,in order to provide reference for the ecological cultivation of A.mongholicus.The results showed that,1)Alpha diversity analysis revealed that with the increase of growth years,the Observed species index and Chao1 index of the rhizosphere soil bacterial community of A.mongholicus showed a trend of first decreasing and then increasing,while the Shannon index and Simpson index showed little change trend.Beta diversity analysis revealed that at the genus level,there were 47,26,and 12 differential species in H1-vs-HCK,H2-vs-HCK,and H1-vs-H2.The predicted functional genes were enriched in KEGG pathways such as carotenoid biosynthesis,glycerolipid metabolism,and glycerophoripip metabolism,etc,respectively.2)Metabolomics analysis revealed there were 538,225,and 559 differential metabolites in H1-vs-HCK,H2-vs-HCK,and H1-vs-H2.These metabolites were enriched in 128,70,and 150 KEGG pathways.Correlation analysis revealed that the certain differential species relative abundance were significantly correlated with the differential metabolites expression levels.3)Enzyme activity analysis revealed that increased growth years reduced the catalase,alkaline phosphatase,and urease activities in the rhizosphere soil,while increasing the sucrase activity.Correlation analysis found a highly significant correlation between the bacterial community diversity and the certain enzymes activity.In summary,A.mongholicus seeds and seedlings have different effects on the bacterial community diversity,metabolites,and enzyme activity of rhizosphere soil during their growth and development.
