以37份黑木耳(Auricularia heimuer)栽培菌株为试材,采用苯胺蓝平板褪色试验、愈创木酚平板显色试验、拮抗试验、不同温度(15、20、25 ℃)培养及液体发酵胞内外多糖含量、菌丝体干物质含量测定等方法,研究了不同菌株产木质素降解酶能力、遗传差异、菌丝生长速率、胞内外多糖产量及菌丝生物量等指标,以期为黑木耳的深度开发利用提供参考依据.结果表明:通过苯胺蓝和愈创木酚平板筛选获得23株产酶能力较强的菌株;拮抗试验进一步分析了 23份菌株的遗传多样性;菌丝生长速率随温度升高显著提高,其中AH33('菊3')在20 ℃和25 ℃下生长速率最快;多糖产量方面,菌株AH17('新科10号')胞外多糖产量最高(27.316 mg·mL-1),AH22('牡丹1号')胞内多糖产量最高(60.193 mg·mL-1);相关性分析表明,发酵菌丝生物量与15 ℃菌丝生长速度呈极显著正相关,与20、25 ℃生长速度、胞内外多糖含量均呈负相关;综合分析表明,菌株AH17('新科10号')在生长适应性、多糖产量及生物量等方面表现优异,是最具应用潜力的优势菌株,推荐作为后续高产栽培或发酵生产的候选材料.

Taking 37 cultivated Auricularia heimuer strains as test materials,aniline blue plate decolorization tests,guaiacol plate chromogenic tests,antagonism tests,culture at different temperatures(15 ℃,20 ℃,and 25 ℃),and submerged fermentation combined with determination of extracellular and intracellular polysaccharide content and mycelial dry matter content were employed to investigate lignin-degrading enzyme production capacity,genetic differences,mycelial growth rate,extracellular and intracellular polysaccharide yield,and mycelial biomass of different strains,in order to provide reference for Auricularia heimuer depth development and utilization.The results showed that 23 strains with strong enzyme-producing ability were obtained through aniline blue and guaiacol plate screening;antagonism tests further analyzed the genetic diversity of these 23 strains.Mycelial growth rate increased significantly with temperature,with AH33('Ju 3')exhibiting the fastest growth rate at 20 ℃ and 25 ℃.Regarding polysaccharide yield,strain AH17('Xinke 10')had the highest extracellular polysaccharide yield(27.316 mg·mL-1),while AH22('Mudan 1')had the highest intracellular polysaccharide yield(60.193 mg·mL-1).Correlation analysis showed that fermentation mycelial biomass was extremely significantly positively correlated with mycelial growth rate at 15 ℃,but negatively correlated with growth rates at 20 ℃ and 25 ℃,as well as with intracellular and extracellular polysaccharide content.Comprehensive analysis indicated that strain AH17('Xinke 10')exhibited excellent performance in growth adaptability,polysaccharide yield,and biomass,making it the most promising candidate strain and recommended for subsequent high-yield cultivation or fermentation production.
