Optimization of Extraction Process for Seven Phenolic Acids and Flavonoids from Young Fruits of Chaenomeles speciosa Nakai Based on the Box-Behnken Design

目的 优化宣木瓜幼果中酚酸及黄酮类7种成分的提取工艺.方法 以宣木瓜幼果为研究对象,采用高效液相色谱法同时测定莽草酸、苹果酸、没食子酸、绿原酸、原花青素B1、原花青素B2、儿茶素共7种成分的含量;在单因素试验基础上筛选关键影响因素,结合Box-Behnken响应面设计,以7种成分的总含量为评价指标,优化提取工艺参数.结果 获得的最佳提取工艺条件:提取溶剂为0.3%磷酸溶液,料液比1∶30 g/mL,提取时间30 min.以此工艺进行5组验证试验,测得7种成分总含量的实际值为47.136 mg,与预测值46.707 mg接近.各因素对总提取率的影响顺序由大到小依次为提取时间、料液比、提取溶剂.结论 所建立的模型预测性良好,优化后的宣木瓜幼果提取工艺稳定可行.

Objective To deeply optimize the extraction process for seven phenolic acids and flavonoids from young fruits of Chaenomeles speciosa Nakai.Methods With the young fruits of Chaenomeles speciosa Nakai as the research object,high-performance liquid chromatog-raphy was used for simultaneous determination of shikimic acid,malic acid,gallic acid,chlorogenic acid,proanthocyanidin B1,proanthocya-nidin B2,and catechin.Key influencing factors were screened based on single-factor experiments,and the Box-Behnken response surface design was used to optimize extraction process parameters with the total content of the seven components as the assessment index.Results The optimal extraction process conditions obtained were 0.3%phosphoric acid solution as the extraction solvent,a solid-liquid ra-tio of 1∶30 g/mL,and an extraction time of 30 minutes.Five validation experiments were conducted under these conditions,and the actual total content of the seven components was 47.136 mg,which was close to the predictive value of 46.707 mg.Extraction time had the grea-test influence on total extraction rate,followed by solid-liquid ratio and extraction solvent.Conclusion The model established has good predictability,and the optimized extraction process for young fruits of Chaenomeles speciosa Nakai is stable and feasible.
