地构叶治疗类风湿关节炎的潜在活性物质研究OA

Potential Active Substances in Speranskia tuberculata for Treatment of Rheumatoid Arthritis

目的 探究地构叶治疗类风湿关节炎的潜在活性物质和可能机制.方法 采用皮下注射牛Ⅱ型胶原与弗氏佐剂的混合液复制小鼠类风湿关节炎模型;将小鼠随机分为空白组,模型组,雷公藤多苷片组,地构叶石油醚部位高、低剂量组,地构叶乙酸乙酯部位高、低剂量组,地构叶正丁醇部位高、低剂量组和地构叶水部位高、低剂量组,观察地构叶提取物对类风湿关节炎的影响;采用网络药理学方法获取地构叶的潜在活性成分及靶点,并结合分子对接,模拟核心靶点和成分的对接模式以揭示地构叶的作用机制.结果 地构叶乙酸乙酯部位高、低剂量和地构叶水部位高、低剂量均能显著降低关节炎评分,抑制滑膜组织增生,减少炎症细胞浸润,并降低血清炎症因子白细胞介素-6、白细胞介素-1β和肿瘤坏死因子-α水平.地构叶所含秦皮乙素与大波斯菊苷等活性成分,可能通过作用于肿瘤坏死因子、TP53、AKT1等关键靶点,进而影响内分泌耐药性、MAPK信号通路及癌症中前列腺素代谢等在类风湿关节炎发病机制中起重要作用的相关通路.结论 地构叶提取物对胶原诱导的类风湿关节炎具有明显的改善作用,其活性物质可能为秦皮乙素与大波斯菊苷等,这些成分通过多靶点、多通路的方式发挥治疗作用.

Objective To investigate the potential active substances in Speranskia tuberculata for the treatment of rheumatoid arthritis and their possible mechanisms.Methods A mouse model of rheumatoid arthritis was established by subcutaneous in-jection of a mixture of bovine type Ⅱ collagen and complete Freund's adjuvant,and the mice were randomly divided into blank group,model group.Tripterygium glycosides tablet group,high-/low-dose Speranskia tuberculata-petroleum ether site groups,high-/low-dose Speranskia tuberculata-ethyl acetate site(ST-EAC)groups,high-/low-dose Speranskia tuberculata-n-butanol site groups,and high-/low-dose Speranskia tuberculata-water site(ST-W)groups.The effect of Speranskia tubercu-lata extracts on rheumatoid arthritis was observed.The method of network pharmacology was used to obtain the potential ac-tive components of Speranskia tuberculata and their targets,and molecular docking was also performed to simulate the docking patterns between core targets and components and reveal the mechanism of action of Speranskia tuberculata.Results The high-and low-dose ST-EAC and the high-and low-dose ST-W significantly reduced arthritis scores,inhibited synovial hyper-plasia,alleviated inflammatory cell infiltration,and reduced the serum levels of interleukin-6,interleukin-1β,and tumor necro-sis factor-α.The active components of Speranskia tuberculata,such as aesculetin and apigenin-7-O-glucoside,might act on the key targets such as TNF,TP53,and AKT1,thereby affecting the important pathways in the pathogenesis of rheumatoid ar-thritis including endocrine resistance,MAPK,and prostaglandins in cancer.Conclusion The extract of Speranskia tubercula-ta can significantly improve collagen-induced rheumatoid arthritis,and its active substances aesculetin and apigenin-7-O-gluco-side can exert a therapeutic effect through multiple targets and pathways.
