[目的]探明不同生长年限兰州百合根际土壤理化特征和细菌群落结构,为兰州百合根际土壤微环境的构建和保持提供理论依据.[方法]以4年(G4)、6年(G6)和8年(G8)的兰州百合根际土壤为研究对象,分别测定其土壤理化指标,同时利用高通量测序技术分析土壤细菌群落特征.[结果]随着兰州百合生长年限的增加,根际土壤pH 逐渐降低,有机质、碱解氮整体呈上升趋势,有效磷、速效钾呈先增大后减小趋势.硝酸还原酶、脲酶、β-葡萄糖苷酶和过氧化氢酶活性均显著高于 CK 土壤(P<0.05).碱性磷酸酶活性随着百合生长年限的增加而增大.G8土壤多酚氧化酶活性显著高于G4和G6(P<0.05).细菌多样性结果表明,不同生长年限兰州百合土壤的优势门类主要是变形杆菌门(Proteobacteria)、酸杆菌门(Acidobacteriota)、拟杆菌门(Bacteroidota)、放线菌门(Actinobacteriota)和绿弯菌门(Chloroflexi),相对丰度在 88%以上.变形杆菌门和酸杆菌门相对丰度较高,不同生长年限间无显著差异(P>0.05),拟杆菌门和放线菌门,拟杆菌门在G4土壤中丰度最高,为16.02%,在G6和G8 中分别为13.10%和14.96%,显著高于 CK 的6.29%.而放线菌门在G6中丰度最高为12.43%,在G4中为10.10%,在G8中降低至8.16%,与CK 无显著差异.[结论]兰州百合根际土壤理化性质和细菌群落结构随着百合生长年限增加发生了明显变异,其中土壤pH、碱性磷酸酶、多酚氧化酶、拟杆菌门和放线菌门等关键指标有利于维持根际土壤微环境稳定.该研究结果可为兰州百合优质高产栽培提供理论依据.

[Objective]To explore the physicochemical characteristics and bacterial community structure of the rhizosphere soil of Lanzhou lilies with different growth years,and provide theoretical basis for the construction and maintenance of the rhizosphere soil microenvironment of Lanzhou lilies.[Method]This study focused on the rhizosphere soil of Lanzhou lilies with growth years of 4(G4),6(G6),and 8(G8)years,and measured their soil physicochemical indicators.At the same time,high-throughput sequencing technology was used to analyze the charac-teristics of soil bacterial communities.[Result]The results showed that as the growth years of Lanzhou lily increased,the pH value of the rhi-zosphere soil gradually decreased,and the organic matter and alkaline nitrogen showed a U-shaped change.The available phosphorus and a-vailable potassium first increased and then decreased.The content of nitrate reductase,urease,β-glucosidase and catalase showed a U-shaped change overall,but both were significantly higher than those in the CK group soil(P<0.05).The content of alkaline phosphatase increased with the growth years of Lanzhou lily.The content of polyphenol oxidase in G8 soil was significantly higher than that in G4 and G6(P<0.05).The results of bacterial diversity showed that the dominant phyla of Lanzhou lily soil with different growth years were Proteobacteria,Acidobac-teriota,Bacteroidota,Actinobacteriota,and Chloroflexi,with relative abundance exceeding 88%.The relative abundance of Proteobacteria and Acidobacteria was the highest,and there was no significant change between different growth years(P>0.05).The abundance of Bacteroi-dota and Acidobacteriota was the highest in G4 soil at 16.02%,while in G6 and G8 it was 13.10%and 14.96%,respectively,significantly higher than the CK group's 6.29%.The highest abundance of Acidobacteriota was 12.43%in G6,10.10%in G4,and decreased to 8.16%in G8,with no significant difference compared to the CK group.[Conclusion]In summary,the physicochemical and bacterial structure of the rhizosphere soil of Lanzhou lilies vary significantly with the growth years of Lanzhou lily.The regulation of factors such as soil pH,alkaline phosphatase,polyphenol oxidase,Bacteroidota,and Acidobacteriota with significant changes is beneficial for maintaining the healthy and sta-ble rhizosphere soil microenvironment of Lanzhou lilies.This result may provide a theoretical basis for achieving high-quality and high-yield cultivation of Lanzhou lilies.
