Exploring the targets and mechanism of Qianggu Ning in the treatment of postmenopausal osteoporosisOA

Background:Clinical observations have indicated that Qianggu Ning(QGN)can enhance bone density in postmenopausal women.We aimed to identify the primary active ingredients and targets of via network pharmacology and biological validation while exploring its potential mechanisms of action.Methods:We employed ultra-high-performance liquid chromatography-mass spectrometry to detect and analyze the active components of the QGN decoction.Topological analysis and target prediction were conducted in conjunction with network pharmacology.The binding efficiency of target proteins and small molecular compounds was verified using molecular docking technology.We simulated a mice model of postmenopausal osteoporosis(PMOP)through bilateral bilateral ovariectomy and assessed bone density changes in ovariectomized(OVX)mice after QGN intervention.We validated biological functions and target genes via pathological testing and quantitative polymerase chain reaction.Results:QGN effectively mitigates bone loss and lipid metabolism disorders in OVX mice.Using network pharmacology,we predicted that the key target genes for alleviating PMOP with QGN.The principal components of QGN underwent molecular docking with these five target genes,all demonstrating robust binding activity.Conclusion:This study confirms the efficacy of QGN in treating PMOP.QGN may achieve the desired effect of increasing bone density via pathways related to lipid,atherosclerosis and MAPK signaling.
