Current research status on the role of natural product polyphenols in anti-liver injuryOA

Liver injury is a widespread global health concern characterized by a complex etiology and limited therapeutic alternatives,underscoring the urgent need for novel hepatoprotective agents.Natural polyphenolic compounds,including phenolic acids,flavonoids,and stilbenes,are abundantly found in traditional Chinese medicine and various natural products.In recent years,polyphenols have shown remarkable potential in ameliorating liver injury and associated disorders,owing to their potent antioxidant,anti-inflammatory,and metabolic regulatory properties.Although existing reviews have examined the hepatoprotective effects of polyphenols,systematic studies that comprehensively classify polyphenols derived from plants,animals,and fungi remain insufficient.This review elucidates the mechanisms by which natural polyphenols mitigate liver injury,focusing on their protective effects through the regulation of inflammatory responses,oxidative stress,mitochondrial dysfunction,apoptosis,and autophagy.It also analyzes the relationship between the structural diversity of polyphenols(e.g.,catechol structures,stilbene derivatives such as resveratrol,and phenolic acids)and their biological activity.Furthermore,this review highlights the emerging role of the gut-liver axis in mediating the effects of polyphenol metabolites,while acknowledging the limitations in current research regarding bioavailability,clinical translation,and the synergistic effects of polyphenols.Future research directions are proposed to provide theoretical foundations and practical references for the development of innovative hepatoprotective drugs.
