Evaluation of the differences in Radix Glycyrrhizae quality among different planting methods based on quantitative chemistry
通过探究仿野生种植与田间种植方式下甘草特征指标的差异,综合评价不同种植方式甘草质量差异,为人工规范化种植甘草提供技术指导.运用中药材显微鉴别(MI)、薄层色谱鉴别(TLC)、高效液相色谱(HPLC)含量测定等技术手段,结合主成分分析(PCA)和聚类分析(HCA)化学计量学方法以及中药色谱指纹图谱相似度评价系统,对不同种植方式下甘草化学活性成分进行分析与比较.MI分析表明,不同种植方式下甘草的显微特征存在一定差异,田间种植品晶纤维和草酸钙方晶显著大于仿野生种植品和空白对照品;PCA和HCA分析表明,不同种植方式下甘草中甘草苷、甘草酸含量存在显著差异;TLC和HPLC指纹图谱分析表明,甘草中甘草酸和甘草苷化学活性成分积累表现为仿野生种植品>田间种植品>空白对照品.因此,科学合理的田间管理技术可有效促进甘草中化学活性成分的积累,提升甘草品质,为甘草规模化、规范化种植提供技术指导,能够进一步提升当地甘草种植产业的竞争力,服务地方经济发展.
By investigating the wild-like cultivation of Radix Glycyrrhizae,the differences in characteristic indicators of Radix Glycyrrhizae under different field planting methods were explored,and the quality differences of Radix Glycyrrhizae under different planting methods were comprehensively evaluated,so as to provide technical guidance for the standardized artificial cultivation of Radix Glycyrrhizae.The chemical component analysis and comparison of Radix Glycyrrhizae under different planting methods were conducted using techniques such as microscopic identification(MI),thin-layer chromatography identification(TLC),high-performance liquid chromatography(HPLC)content determination,combined with chemometric methods such as principal component analysis(PCA)and hierarchical cluster analysis(HCA),as well as the similarity evaluation system of traditional Chinese medicine chromatographic fingerprint.MI analysis indicated that there were certain differences in the microscopic characteristics of Radix Glycyrrhizae under different cultivation methods.The crystall fibers and calcium oxalate prisms of Radix Glycyrrhizae planted in the field were significantly larger than those of wild-like cultivation and the blank control.PCA and HCA analysis showed that there were significant differences in the contents of glycyrrhizin and glycyrrhonic acid in Radix Glycyrrhizae under different cultivation methods.TLC and HPLC fingerprint analysis indicated that the accumulation of chemical active components such as glycyrrhizic acid and glycyrrhizin in Radix Glycyrrhizae was in the order of wild-like cultivation>field planting>blank control.Therefore,scientific and reasonable field management techniques can effectively promote the accumulation of chemical active components in Radix Glycyrrhizae,improve the quality of Radix Glycyrrhizae,provide technical guidance for the large-scale and standardized cultivation of Radix Glycyrrhizae,and further enhance the competitiveness of Radix Glycyrrhizae planting industry in the local area,serving local economic development.
