Antioxidant and immunoenhancing activities of polysaccharides from Panax quinquefolium L.root hairy in vitro

为探究西洋参须多糖的体外生物活性,本研究采用不同浓度乙醇分级沉淀法制备了5种西洋参须多糖组分(PQLP40、PQLP50、PQLP60、PQLP70、PQLP80),并对其体外免疫增强活性及体外抗氧化活性进行评价.通过MTT法测定各组分对RAW 264.7巨噬细胞增殖的影响,评估西洋参须多糖的免疫活性;通过测定各组分对DPPH·、·OH、·O2-和ABTS+·等自由基的清除活性,评估西洋参须多糖的体外抗氧化活性.结果显示,PQLP40(62.50 µg/mL)和PQLP70(31.25 µg/mL、125.00 µg/mL、500.00µg/mL)均可显著促进RAW 264.7细胞增殖.在抗氧化活性方面,不同组分多糖表现出不同的自由基清除优势,PQLP80对DPPH·和·OH的清除活性最强,PQLP40对·O2-的清除活性最强,PQLP60对ABTS+·的清除活性最强.研究表明,醇沉浓度可影响西洋参须多糖体外免疫活性和抗氧化活性,且不同醇沉浓度的多糖之间的免疫活性和抗氧化活性存在差异,其中PQLP40具有较强的免疫增强活性,PQLP80具有较强的抗氧化活性.

To investigate the in vitro biological activity of polysaccharides from Panax quinquefolium L.root hairy,five polysaccharide components(PQLP40,PQLP50,PQLP60,PQLP70,PQLP80)were prepared by using different concentrations of ethanol fractionation precipitation,and in vitro immune enhancement and antioxidant activities were evaluated.The effect of each component on the proliferation of RAW 264.7 macrophages was determined by the MTT assay to assess its immune activity;its antioxidant activity was evaluated by measuring its scavenging ability against DPPH·radicals,hydroxyl radicals,superoxide anion radicals,and ABTS+·cation radicals.In terms of immunological activity,both PQLP40(62.50 µg/mL)and PQLP70(31.25 µg/mL,125.00 µg/mL,500.00 µg/mL)could significantly promote the proliferation of RAW 264.7 cells.Regarding antioxidant activity,polysaccharides from different fractions exhibited different free radical scavenging strengths:PQLP80 had the strongest scavenging activity for DPPH·and·OH radicals,PQLP40 was most effective against·O2-radicals,and PQLP60 showed the highest scavenging activity for ABTS+·.The study indicates that the alcohol precipitation concentration affects the in vitro immunomodulatory and antioxidant activities of Panax quinquefolius polysaccharides,and there are differences in activity among polysaccharides obtained at different alcohol concentrations.Among them,PQLP40 and PQLP80 demonstrate the best immuno-antioxidant activities.
