本研究旨在探讨多房棘球绦虫(Echinococcus multilocularis)干细胞相关基因Nanos-1/2(Em-Nos1/2)的分子特征、蛋白表达及Nos蛋白在虫体中的定位.对Em-Nos1/2基因进行生物信息学分析,通过构建原核表达载体pET28a-Em-Nos1/2,诱导表达并纯化重组蛋白,免疫家兔制备多克隆抗体;利用Western blot和免疫荧光技术分别检测抗体的特异性及天然蛋白在虫体中的定位.多房棘球蚴Em-Nos1/2系统进化分析显示,这两个基因在绦虫属中呈现高度保守性,但三维结构存在细微差异.构建原核表达载体pET28a-Em-Nos1/2,经诱导表达与蛋白纯化后,获得高纯度的重组Em-Nos1/2蛋白,并制备兔多克隆抗体,其效价可达1∶204 800.Western blot试验结果证实,所制备的多克隆抗体可特异性识别多房棘球绦虫的天然Em-Nos1/2蛋白.免疫荧光定位结果揭示,Em-Nos1/2蛋白可特异性表达于多房棘球蚴生发层.本研究成功制备了能特异性识别天然Em-Nos1/2蛋白的多克隆抗体,并证实Nos蛋白定位于多房棘球蚴生发层,为进一步探究Em-Nos1/2基因在多房棘球蚴干细胞中的生物学功能提供了关键工具和重要基础.

The objective was to investigate the molecular characteristics,protein expression,and localization of the stem cell-associated genes Nanos-1/2 of Echinococcus multilocularis(Em-Nos1/2)in the parasite.Bioinformatic analysis of Em-Nos1/2 was performed.Prokaryotic expression vectors pET28a-Em-Nos1/2 were constructed,and the recombinant proteins were induced,expressed,and purified.Polyclonal antibodies were generated by immunizing rabbits with the purified recombinant proteins.Western blot and immunofluorescence assays were employed to assess antibody specificity and determine the localization of the native proteins in the parasite,respectively.Phylogenetic analysis of Em-Nos1/2 from E.multilocularis revealed that these two genes were highly conserved within the genus Echinococcus,although minor differences in their three-dimensional structures were observed.High-purity recombinant Em-Nos1/2 proteins were obtained following construction of pET28a-Em-Nos1/2 vectors,induction of expression,and protein purification.The generated rabbit polyclonal antibodies exhibited high titers,reaching up to 1∶204 800.Western blot analysis confirmed that the prepared polyclonal antibodies specifically recognized the native Em-Nos1/2 proteins from E.multilocularis.Immunofluorescence localization demonstrated that both Em-Nos1/2 proteins were specifically expressed in the germinal layer of E.multilocularis larvae(metacestode).In this study,polyclonal antibodies capable of specifically recognizing the native Em-Nos1/2 proteins were successfully generated,and their specific localization to the germinal layer of E.multilocularis metacestodes was confirmed.These findings can provide crucial tools and a foundational basis for further investigations into the biological functions of Em-Nos1/2 in the stem cells of E.multilocularis.
