177份花生种质资源种仁与荚壳的黄曲霉侵染抗性鉴定与评价OA
Identification and Evaluation for Resistance to Aspergillus flavus Infection in Seeds and Shells of 177 Arachis hypogaea Germplasm Resources
花生黄曲霉毒素污染直接威胁花生产业的健康可持续发展,显著制约我国花生出口贸易.为筛选种仁或荚壳抗黄曲霉侵染的特异性花生种质资源,同时发掘兼具种仁-荚壳双重抗性的优异材料,采用孢子菌悬液侵染法,以177份花生种质为试验材料,开展种仁和荚壳的黄曲霉侵染抗性系统鉴定.结果表明,参试花生材料种仁的侵染指数分布范围为1.67%~96.33%,其中抗性种质6份、中抗48份、中感86份、感病33份、高感4份,中抗以上材料占比30.51%;荚壳抗性试验筛选出高抗材料5份、中抗55份、中感102份、高感15份,中抗以上材料占比33.9%;其中'花育60'表现为种仁抗、荚壳高抗,是所有试验材料中唯一兼具种仁和荚壳双重抗性的优异种质;相关性分析表明,花生荚壳侵染指数与种仁侵染指数之间无显著相关性.共筛选出6份种仁抗性、5份荚壳高抗性和1份双重抗性的花生抗黄曲霉优异种质,为黄曲霉抗性基因挖掘和抗黄曲霉花生品种的遗传改良提供了关键基础材料.
The contamination of Arachis hypogaea with aflatoxin poses a direct threat to the healthy and sustainable development of the Arachis hypogaea industry and significantly restricts China's Arachis hypogaea export trade.To screen specific Arachis hypogaea germplasm resources that are resistant to Aspergillus flavus infection in the seed kernels or pod shells,and to discover excellent materials with dual resistance in both seed kernels and pod shells,the spore fungus suspension infec-tion method was adopted.With 177 Aspergillus flavus germplasm materials as the experimental materials,a systematic identification of Aspergillus flavus infection resistance in seed kernels and pod shells was conducted.The results showed that the infection index of the tested Arachis hypogaea ma-terials in the seed kernels ranged from 1.67 to 96.33,among which 6 were resistant,48 were moder-ately resistant,86 were moderately susceptible,33 were highly susceptible and 4 were highly highly susceptible.The proportion of materials with resistance above the medium level was 30.51%.In the pod shell resistance test,5 highly resistant materials,55 moderately resistant materials,102 moder-ately susceptible materials,and 15 highly susceptible materials were screened out.Among them,Huayu60 showed resistance in the seed kernels and high resistance in the pod shells,being the only excellent germplasm with dual resistance in both seed kernels and pod shells in this study.Correlation analysis indicated that there was no significant correlation between the infection index of Arachis hy-pogaea pods and that of the seed kernels.A total of 6 Arachis hypogaea materials with resistance in the seed kernels,5 highly resistant materials in the pod shells,and 1 material with dual resistance were screened out as excellent Arachis hypogaea materials resistant to Aspergillus flavus.This pro-vided key basic materials for the mining of Aspergillus flavus resistance genes and the genetic improvement of aflatoxin-resistant Arachis hypogaea varieties.
