目的 鉴定鲜地黄在血热出血模型大鼠体内的代谢产物.方法 16只大鼠随机分为2组,分别灌胃给予生理盐水、鲜地黄提取液,超高效液相色谱-线性离子阱串联静电场轨道高分辨质谱(UHPLC-LTQ-Orbitrap-MS/MS)法鉴定血清、尿液、组织(心、肝、脾、肺、肾、小肠)中原型成分及代谢产物.结果 共鉴定出51种成分,包括6种原型成分、45种代谢产物.在血清、尿液、心、肝、脾、肺、肾、小肠中,分别鉴定出原型成分5、6、1、1、1、1、1、4种,代谢产物9、30、8、11、5、4、6、11种.鲜地黄主要通过脱氢、还原、脱水、甲基化、葡萄糖醛酸化等途径代谢.结论 鲜地黄在血热出血模型大鼠体内发生Ⅰ相、Ⅱ相代谢,代谢产物广泛分布于各主要器官中,可为该药材药效物质基础、作用机制研究提供依据.

AIM To identify the in vivo metabolites of fresh Rehmanniae Radix in blood heat and hemorrhage model rats.METHODS Sixteen rats were randomly divided into two groups and given intragastric administration of normal saline and fresh Rehmanniae Radix extract,respectively,after which ultra-high-performance liquid chromatography coupled with linear ion trap-tandem electrostatic field orbital high-resolution mass spectrometry(UHPLC-LTQ-Orbitrap-MS/MS)was adopted in the identification of prototypical components and metabolites in serum,urine and tissues(heart,liver,spleen,lung,kidney,small intestine).RESULTS Total 51 constituents were identified,including 6 prototypical components and 45 metabolites.In the serum,urine,heart,liver,spleen,lung,kidney,small intestine,5,6,1,1,1,1,1,1,1,4 prototypical components and 9,30,8,11,5,4,6,11 metabolites were identified,respectively.Fresh Rehmanniae Radix was metabolized by pathways such as dehydrogenation,reduction,dehydration,methylation and glucuronidation.CONCLUSION Fresh Rehmanniae Radix occurrs phase Ⅰ and phase Ⅱ metabolisms in blood heat and hemorrhage model rats in vivo,the metabolites are widely distributed in various main organs,which can provide basis for the research on pharmacodynamic material basis and action mechanisms of this medicinal material.
