基于HPLC指纹图谱、化学模式识别和含量测定评价黄芩质量OA
Quality evaluation of Scutellaria baicalensis based on HPLC fingerprints,chemical pattern recognition and content determination
目的 评价黄芩质量.方法 建立HPLC指纹图谱,进行聚类分析、主成分分析.以黄芩苷为内标,计算野黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、千层纸素A、千层纸素A苷相对校正因子,一测多评法测定其含量.结果 30批药材指纹图谱中有15个共有峰,鉴定出7个,相似度0.821~0.999.各批药材聚为2类,3个主成分累积方差贡献率为86.049%.7种成分在各自范围内线性关系良好(r ≥ 0.999 7),平均加样回收率93.02%~99.85%,RSD 1.12%~1.98%,一测多评法、外标法所得结果接近.结论 该方法稳定可靠,可用于黄芩的质量控制.
AIM To evaluate the quality of Scutellaria baicalensis Georgi.METHODS The HPLC fingerprints were established,after which cluster analysis and principal component analysis were performed.Baicalin was used as an internal standard to calculate the relative correction factors of scutellarin,wogonoside,baicalein,wogonin,oroxylin A and oroxin A,then their contents were determined by quantitative analysis of multi-components by single-marker(QAMS)method.RESULTS There were 15 common peaks in the fingerprints for 30 batches of medicinal materials with the similarities of 0.821-0.999,7 of which were identified.Various batches of medicinal materials were clustered into two catagories,three principal components demonstrated the accumulative variance contribution rate of 86.049%.Seven constituents showed good linear relationships within their own ranges(r≥0.999 7),whose average recoveries were 93.02%-99.85%with the RSDs of 1.12%-1.98%.The result obtained by QAMS approximated those obtained by external standard method.CONCLUSION This stable and reliable method can be used for the quality control of S.baicalensis.
