为研究青霉素V钾可溶性粉对由产气荚膜梭菌引起鸡坏死性肠炎(NE)的治疗作用,并确定合理的给药剂量,本试验将250羽由产气荚膜梭菌自然感染引起NE的病鸡随机分为5个组,分别为高剂量(HDG)组、中剂量(MDG)组、低剂量(LDG)组、阴性对照(NCG)组和阳性药物对照(LHS)组,HDG、MDG和LDG组试验鸡分别口服给予102、68和46 mg/(kg·bw)青霉素V钾可溶性粉,LHS组给予盐酸林可霉素可溶性粉,1.5 g/1 L水,混饮,每日1次,连用5 d;NCG组给予等量生理盐水.给药前(D0)、给药第5天(D5)、给药后第7(D12)和14天(D19)对各组试验鸡进行临床观察,对其精神状态、采食状况和腹泻症状进行评分,同时测定体重、采食量和饲料转化率,并采集肛拭子和肠道组织样本进行产气荚膜梭菌的分离和鉴定;试验期间对病死鸡进行剖检和病理学观察;试验结束后进行疗效评价.结果显示,各试验组鸡均表现出不同程度的精神沉郁、食欲减退和腹泻症状;与NCG组相比,LDG、MDG、HDG和LHS组在D5、D12和D19时试验鸡的精神状态评分、采食状况评分和腹泻症状评分均显著降低(P<0.05),体重均显著增加(P<0.05),在D0~D19时试验鸡采食量均显著增加(P<0.05),试验期间各组试验鸡的饲料转化率以NCG组在D1~D5时最高,为4.73.LDG、MDG、HDG和LHS组在D5、D12和D19时产气荚膜梭菌的检出率较NCG组明显降低.试验期间,NCG组死亡7羽,LDG组死亡6羽,HDG组死亡1羽,病死鸡剖检可见NE典型病变.LDG、MDG、HDG和LHS组综合治愈率分别为62%、86%、86%和86%.结果表明,青霉素V钾可溶性粉以68 mg/(kg·bw)剂量临床给药,可有效治疗由产气荚膜梭菌引起的NE,本试验结果可为临床提供治疗依据.

This study aimed to evaluate the therapeutic efficacy of penicillin V potassium soluble powder against necrotic enteritis(NE)in chickens caused by Clostridium perfringens and to determine an appropriate dosage regimen.A total of 250 chickens naturally infected with NE were randomly divided into five groups:high-dose group(HDG),medium-dose group(MDG),low-dose group(LDG),negative control group(NCG),and positive drug control group(LHS).Chickens in the HDG,MDG,and LDG groups were orally administered penicillin V potassium soluble powder at doses of 102,68,and 46 mg/(kg·bw),respectively.Chickens in the LHS group received lincomycin hydrochloride soluble powder(1.5 g/1 L drinking water),administered once daily for 5 consecutive days.Chickens in the NCG group were given an equivalent volume of physiological saline.Clinical observations were conducted before treatment(D0),on day 5 of treatment(D5),and on days 7(D12)and 14(D19)after treatment.The mental status,feed intake,and diarrhea symptoms of chickens were scored.Body weight,feed intake,and feed conversion ratio were recorded.Cloacal swabs and intestinal tissue samples were collected for isolation and identification of C.perfringens.During the experiment,dead chickens were necropsied and subjected to pathological examination.At the end of the trial,treatment efficacy was evaluated.The results showed that chickens in all groups exhibited varying degrees of depression,reduced feed intake,and diarrhea.Compared with the NCG group,the LDG,MDG,HDG,and LHS groups showed significantly lower scores for mental status,feeding behavior,and diarrhea symptoms at D5,D12,and D19(P<0.05),and significantly increased body weight(P<0.05).Feed intake during D0-D19 was significantly increased in the treatment groups compared with the NCG group(P<0.05).The highest feed conversion ratio during D1-D5 was observed in the NCG group(4.73).The detection rate of C.perfringens in the LDG,MDG,HDG,and LHS groups at D5,D12,and D19 was markedly lower than that in the NCG group.During the experiment,7 chickens died in the NCG group,6 in the LDG group,and 1 in the HDG group.Necropsy of the dead chickens revealed typical lesions of NE.The overall cure rates in the LDG,MDG,HDG,and LHS groups were 62%,86%,86%,and 86%,respectively.These results indicate that penicillin V potassium soluble powder at a dose of 68 mg/(kg·bw)is an effective clinical treatment for NE caused by C.perfringens.The findings provide treatment evidence to support its clinical application.
