为了探究牛奶血清淀粉样蛋白A3(SAA3)含量与中红外光谱(MIR)数据之间的关联性,本试验采集244份牛奶样本,采用酶联免疫吸附试验(ELISA)定量测定牛奶SAA3含量,并利用CombiFoss FT+光谱仪获取波段5 011~925 cm-1范围内的1 060个光谱波数的透射率.基于Shapiro-Wilk检验法评估数据的分布特征后,使用Spearman相关性分析计算各光谱波数与牛奶SAA3含量的相关系数.结果显示,牛奶样本的SAA3含量介于0.2~1.6 μg/mL,平均含量为(0.65±0.21)μg/mL.MIR按光谱特征划分为5个子区域,分别为短波红外区(SWIR,5 011~3 673 cm-1)、短中波红外过渡区(SWIR~MWIR,3 669~3 052 cm-1)、中波红外区1(MWIR-1,3 048~1 701 cm-1)、中波红外区2(MWIR-2,1 698~1 586 cm-1)和中长波红外过渡区(MWIR~LWIR,1 582~925 cm-1),其中,SWIR~MWIR和MWIR-2区域均表现较高的光谱变异程度.相关性分析结果显示,43个波数与牛奶SAA3含量的相关性相对较高,相关系数绝对值超过0.1,其中,16个波数达到P<0.05水平,主要分布于波段3 155~3 148、1 562~1539和1 466~1 446 cm-1,分别对应蛋白质N-H键振动、酰胺II带吸收以及尿素氮和脂肪代谢相关的光谱特征.本试验鉴定出与牛奶SAA3含量相关的特征光谱波段,为后续探索构建基于机器学习算法的牛奶SAA3含量定量预测模型提供初步的特征波段筛选参考,对推动牛奶MIR技术在奶牛乳房炎临床辅助诊断中的应用提供依据.

To investigate the relationship between milk serum amyloid A3(SAA3)content and mid-infrared(MIR)spectral data,a total of 244 milk samples were collected.Milk SAA3 concentrations were quantified using enzyme-linked immunosorbent assay(ELISA),and transmission spectra were obtained in the 5 011-925 cm⁻¹ range at 1 060 wavenumbers using a CombiFoss FT+spectrometer.Data distribution was assessed using the Shapiro-Wilk test,and Spearman correlation analysis was performed to calculate the correlation coefficients between each spectral wavenumber and milk SAA3 content.The results showed that milk SAA3 concentrations ranged from 0.2 to 1.6 μg/mL,with an average of(0.65±0.21)μg/mL.The MIR spectrum was divided into five sub-regions based on spectral characteristics:short-wave infrared(SWIR,5 011-3 673 cm⁻¹),short-mid-wave infrared transition(SWIR-MWIR,3 669-3 052 cm⁻¹),mid-wave infrared 1(MWIR-1,3 048-1 701 cm⁻¹),mid-wave infrared 2(MWIR-2,1 698-1 586 cm ⁻¹),and mid-longwave infrared transition(MWIR-LWIR,1 582-925 cm ⁻¹).The SWIR-MWIR and MWIR-2 regions exhibited relatively high spectral variability.Correlation analysis revealed that 43 wavenumbers had relatively high correlations with milk SAA3 content,with absolute correlation coefficients greater than 0.1;among these,16 wavenumbers reached P<0.05,primarily distributed in the 3 155-3 148,1 562-1 539,and 1 466-1 446 cm⁻¹ regions,corresponding to N-H bond vibrations of proteins,amide II band absorption,and spectral features related to urea nitrogen and lipid metabolism,respectively.This study identified MIR spectral bands associated with milk SAA3 content,providing preliminary reference for selecting characteristic wavelengths for future quantitative prediction models based on machine learning.These findings also support the potential application of MIR spectroscopy in the clinical auxiliary diagnosis of bovine mastitis.
