浙江省某鸡场火鸡组织滴虫和异刺线虫感染调查和分析OA
Investigation and Analysis of Histomonas meleagridis and Heterakis gallinarum Infections in a Poultry Farm of Zhejiang Province,China
为了解浙江省火鸡组织滴虫(H.meleagridis)和异刺线虫(H.gallinarum)的流行情况,本试验对浙江省青田县疑似发生组织滴虫病的某山林散养鸡场展开调查.剖检并观察病鸡;采集粪便和盲肠样品进行显微镜观察和分析;采用聚合酶链式反应(PCR)对样品中H.meleagridis 18S rRNA基因进行扩增和测序;利用M199培养基对H.meleagridis进行体外培养,并对分离株和盲肠内容物进行PCR鉴定比对;基于测序结果进行同源性分析并构建遗传进化树.结果显示,病鸡肝脏表面出现典型的"铜钱斑"样病灶;在粪便和盲肠样品中观察到H.meleagridis鞭毛体、H.gallinarum虫体和虫卵.粪便样品的PCR阳性率为3.4%(4/117),肝脏、粪便、盲肠内容物、盲肠上皮和H.gallinarum虫体样品均可扩增出H.meleagridis 18S rRNA基因特异性条带;通过体外培养成功获得H.meleagridis青田分离株,PCR鉴定结果为阳性.H.meleagridis青田分离株的18S rRNA基因序列与我国安徽分离株AH-2和江苏分离株YZ4的同源性高达99.6%,在遗传进化树上位于同一亚支.结果表明,该鸡场存在H.meleagridis和H.gallinarum混合感染,感染株可能是国内地方虫株.本试验为浙江省H.meleagridis和H.gallinarum混合感染的防控研究提供了参考依据.
To investigate the prevalence of Histomonas meleagridis and Heterakis gallinarum in Zhejiang Province,an epidemiological survey was conducted on a free-range forest poultry farm in Qingtian County suspected of histomoniasis.Necropsy and gross pathological examinations were performed on diseased chickens.Fecal samples and cecal contents were collected for microscopic observation and analysis.Polymerase chain reaction(PCR)was used to amplify and sequence the 18S rRNA gene of H.meleagridis.The parasite was isolated and cultured in vitro using M199 medium,and the isolates were identified and compared with cecal content samples by PCR.Sequence homology analysis and phylogenetic tree construction were conducted based on the obtained sequences.The results showed typical"copper coin spot"like lesions on the liver surface of diseased chickens.Flagellated forms of H.meleagridis,as well as adults and eggs of H.gallinarum,were observed in feces and cecal contents.The PCR-positive rate of fecal samples was 3.4%(4/117),and specific bands of the H.meleagridis 18S rRNA gene were amplified from liver,fecal,cecal content,and parasite samples.A Qingtian isolate of H.meleagridis was successfully obtained through in vitro culture and confirmed by PCR.The 18S rRNA gene sequence of this isolate showed 99.6%homology with the AH-2 strain from Anhui and the YZ4 strain from Jiangsu,and clustered within the same subclade in the phylogenetic tree.These findings indicate a mixed infection of H.meleagridis and H.gallinarum in the investigated farm,and the infecting strains are likely local endemic strains.This study provides a reference for the prevention and control of mixed infections caused by H.meleagridis and H.gallinarum in Zhejiang Province.
