猪流行性腹泻病毒Nsp9突变毒株的构建及生物学特性分析OA
Construction and biological characteristic analysis of a porcine epidemic diarrhea virus Nsp9 mutant viral strain
猪流行性腹泻病毒(PEDV)非结构蛋白Nsp9在病毒复制中具有重要功能,其保守的第59位半胱氨酸参与蛋白在体外溶液中的二聚化及RNA结合.本研究旨在通过构建Nsp9第59位半胱氨酸突变为丙氨酸的重组病毒,探究该位点对PEDV复制能力的影响.利用CRISPR/Cas9介导的反向遗传学技术,以PEDV G2b型流行毒株为骨架,构建Nsp9第59位氨基酸突变的重组病毒.通过间接免疫荧光、Western-blot和病毒生长曲线测定,系统分析该重组病毒的感染性、蛋白表达及复制特性,评估该位点突变对PEDV复制能力的影响.成功获得重组病毒rAH2012/12-Nsp9-C59A,该重组病毒在Vero细胞中可稳定传代,并有效诱导PEDVN蛋白的表达.生长动力学显示,该重组突变株的早期复制趋势与亲本株基本一致,在感染后期病毒滴度高于亲本毒株.本研究揭示了PEDV Nsp9第59位半胱氨酸突变对病毒在Vero细胞中复制的影响,为深入理解PEDV Nsp9的功能提供了新线索.
The nonstructural protein Nsp9 of porcine epidemic diarrhea virus(PEDV)plays an impor-tant role in viral replication,and its conserved cysteine residue at position 59 is involved in protein dimerization and RNA binding in vitro.This study aimed to investigate the impact of this site on PEDV replication capacity by constructing a recombinant virus with a mutation at the 59th amino acid of Nsp9 from cysteine(C)to alanine(A).Using CRISPR/Cas9-mediated reverse genetics technology and based on the backbone of a PEDV G2b epidemic strain,a recombinant virus was constructed with a mutation at the 59th amino acid of Nsp9.Through indirect immunofluorescence,Western-blot,and viral growth curve as-says,the infectivity,protein expression,and replication characteristics of the mutant virus were systematically analyzed to evaluate the impact of this mutation on PEDV replication capacity.The re-combinant virus rAH2012/12-Nsp9-C59A was successfully obtained.It could be stably passaged in Vero cells and effectively induced the expressed of PEDV N protein.Growth kinetics showed that the early replication trend of this recombinant mutant was generally consistent with that of the parental strain,while the viral titer in the later stages of infection was higher than that of the parental strain.This study revealed the impact of the cysteine mutation at position 59 of PEDV Nsp9 on viral replication in Vero cells,providing new insights for a deeper understanding of the function of PEDV Nsp9.
