肺炎支原体R414北京流行株感染小鼠模型的研究OA
Study on a murine model of infection with the Mycoplasma pneumoniae R414 Beijing epidemic strain
目的 以肺炎支原体(Mycoplasma pneumoniae,Mp)模式株FH和北京流行株R414感染BALB/c小鼠建立经鼻感染小鼠Mp模型,并且评估R414株在致病性、免疫病理等方面与模式株FH的异同.方法 3周龄和8周龄BALB/c小鼠滴鼻感染FH株、R414株,对照组用0.9%氯化钠溶液滴鼻,感染后第6、9、12天测定肺组织病原载量、病理及转录组变化、外周血免疫细胞及细胞因子改变.结果 小鼠感染Mp后呈现急性肺炎特征(6 d病原载量高、炎症显著,9~12d减轻),FH株与R414株病原载量无明显差异.感染FH株后,8周龄小鼠的细胞免疫启动早于3周龄;且8周龄小鼠的转录组差异基因数量远超3周龄.R414株感染3周龄小鼠6 d后,外周血IP-10升高,感染8周龄小鼠6 d后,单核细胞占比增多,TNF-α升高.R414株在3周龄小鼠中诱导"病毒应答"通路;8周龄小鼠中则主要激活"IL-17信号通路"和"中性粒细胞趋化"等通路.结论 R414株诱导的宿主免疫应答与模式株之间出现差异化,且与小鼠周龄有相关性,值得进一步研究.
Objective To establish a murine Mycoplasma pneumoniae(Mp)model by intranasally infecting BALB/c mice with the reference strain FH and the Beijing epidemic strain R414,and to evaluate the similarities and differences in pathogenicity and immunopathology between R414 and the reference strain FH.Methods 3-week-old and 8-week-old BALB/c mice were intranasally infected with FH or R414 strains.Control groups received intranasal 0.9%sodium chloride solution.Pathogen load,pathological changes,and transcriptome alterations in lung tissues,as well as peripheral blood immune cell and cytokine levels,were measured on days 6,9,and 12 post-infection.Results Mice infected with Mycoplasma pneumoniae exhibited features of acute pneumonia(high pathogen load and significant inflammation on day 6,subsiding by days 9~12).No significant differences were observed between FH and R414 strains in pathogen load.Following FH infection,cellular immunity in 8-week-old mice was activated earlier than in 3-week-old mice;moreover,the number of differentially expressed genes in the transcriptome of 8-week-old mice significantly exceeded that of 3-week-old mice.6 d after R414 infection in 3-week-old mice,peripheral blood IP-10 levels increased.6 d after R414 infection in 8-week-old mice,the proportion of monocytes increased,and TNF-αlevels rose.The R414 strain induced a"viral response"pathway in 3-week-old mice,while in 8-week-old mice,it primarily activated pathways such as the"IL-17 signaling pathway"and"neutrophil chemotaxis".Conclusions The R414 strain induced differentiated host immune responses compared to the reference strain.These differences were correlated with the age of the mice and warrant further investigation.
