基于"以方测证"的寒湿泄泻大鼠模型建立与验证OA
Establishment and verification of a rat model of cold-dampness diarrhea based on"diagnosis syndrome by prescription"
目的 本研究依托"以方测证"思路,采用内因与外因相结合的方式构建大鼠寒湿泄泻模型.方法 将48只大鼠随机分为空白对照(CON,n=12)组、寒湿泄泻模型(CDS,n=12)组、理中汤治疗寒湿泄泻(LCD,n=12)组及白头翁汤治疗寒湿泄泻(PCD,n=12)组.模型构建采用复合因素:以内因(番泻叶联合冰水灌胃)叠加外因(寒湿环境刺激)复制大鼠寒湿泄泻模型,并以白头翁汤、理中汤干预开展以方测证分析,记录各组大鼠临床表现.电镜下观察组织超微结构变化.检测各组大鼠血清中ATP酶活性及IL-6、IL-10、TNF-α、IL-1β水平.结果 与CON组相比,CDS组大鼠体温和体质量均显著降低(P<0.05);稀便率、粪便含水量和腹泻指数升高,符合寒湿泄泻的主次症辨证诊断标准,CDS组大鼠血清中Na+-K+ATP酶活性、Ca2+-Mg2+ATP 酶活性极显著降低(P<0.05),IL-6 显著升高(P<0.05),IL-1β 极显著升高(P<0.01),IL-10水平显著降低(P<0.05),TNF-α水平升高,但无统计学意义(P>0.05).以方测证结果显示,理中汤对于寒湿泄泻大鼠上述指标具有治疗效果,白头翁汤对于寒湿泄泻大鼠效果不明显.结论 运用"内因+外因"的寒湿泄泻动物模型构建成功,为腹泻类疾病机制及其药物研究提供了良好的方法.
Objective This study is based on the"diagnosis syndrome by prescription"approach and adopts a combination of internal and external factors to construct a rat model of cold-damp diarrhea.Methods 48 rats were randomly divided into a blank control(CON,n=12)group,a cold-damp diarrhea model(CDS,n=12)group,a Lizhong decoction intervention(LCD,n=12)group,and a Pulsatilla decoction intervention(PCD,n=12)group.The model construction used a composite factor:internal factors(psyllium seed combined with ice water gavage)and external factors(cold-damp environmental stimulation)were combined to replicate the rat model of cold-damp diarrhea.The Lizhong decoction and Pulsatilla decoction interventions were carried out for the analysis of the diagnosis syndrome by prescription,and the clinical manifestations of each group of rats were recorded.The atpase activity and the levels of serum interleukin(IL)-6,IL-10,tumor necrosis factors(TNF)-α,and IL-1β were measured in all groups.Results Compared with normal rats,rats in the model group exhibited increased rates of loose stools,higher fecal water content,and elevated diarrhea indices,consistent with the diagnostic criteria for primary and secondary symptoms associated with cold-dampness diarrhea.Notably,serum Ca2+-Mg2+ATPase and Na+-K+ATPase activities were significantly decreased in rats in the model group,while IL-6,IL-1β,and TNF-αlevels were markedly increased and IL-10 levels declined.Prescription testing indicated that Lizhong Decoction had a significant impact on these parameters among rats suffering from cold-dampness diarrhea,while Pulsatilla decoction had no notable effects.Conclusion An effective animal model for cold-dampness diarrhea was successfully established by a"combination of internal causes+external causes",providing a robust methodological foundation for further investigations into the mechanisms underlying diarrhea-related diseases.
