瓜蒌薤白白酒汤通过调节肠道菌群增加NR活性改善小鼠动脉粥样硬化的实验研究OA
Use of Gualou Xiebai Baijiu Decoction for improving atherosclerosis in mice by modulating gut microbiota and increasing nitrate reductase activity
目的 探讨瓜蒌薤白白酒汤(Gualou Xiebai Baijiu Decoction,GXB)能否调节肠道菌群(gut microbiota,GM),促进硝酸盐还原酶(nitrate reductase,NR)活性,改善动脉粥样硬化(atherosclerosis,AS)的作用机制.方法 将SPF级36只8周龄雄性ApoE-/-小鼠随机分为4组:对照(Control)组、模型(Model)组、阿托伐他汀(Ato)组和瓜蒌薤白白酒汤(GXB)组,每组9只小鼠.除Control组外,其余组高脂高胆固醇饲料喂养16周建模后按组别灌胃干预4周.通过油红O及HE染色观察斑块和血管形态,检测血清TG、TC、LDL-C、HDL-C及血清/肠液硝酸盐(nitrate,NO3-)、亚硝酸盐(nitrite,NO2-)、一氧化氮(nitric oxide,NO)水平,并应用宏转录组测序分析GM结构与功能,同时,为验证功能分析的结果,进一步检测血清/肠液中的NR和亚硝酸还原酶(nitrite reductase,NiR)活性.结果 与Model组相比,GXB组显著改善小鼠AS的病理进展,血清TG、TC、LDL-C水平降低,HDL-C升高,血清和肠液中的NO3-、NO2-、NO水平上升.GM分析显示,GXB降低了AS小鼠GM的丰富度、提高均匀度,且显著降低厚壁菌门丰度,回升拟杆菌门和疣微菌门等厌氧菌丰度.功能分析显示,GXB通过促进NR活性,激活NO3--NO2--NO通路,提高了内源性NO水平.此外,与Model组相比,GXB组中血清/肠液的NR、NiR水平升高,与功能分析的结果一致.结论 GXB可能通过改善GM结构与功能,增加肠道厌氧菌的丰度,促进NR的活性,激活NO3--NO2--NO通路,提升内源性NO水平,进而改善血脂紊乱、减轻AS的病理损伤.
Objective To investigate the mechanism by which Gualou Xiebai Baijiu Decoction(GXB)improves atherosclerosis(AS)by modulating gut microbiota(GM),promoting nitrate reductase(NR)activity.Methods Thirty-six male SPF ApoE-/-mice,aged 8 weeks,were divided randomly into Control,Model,Atorvastatin(Ato),and Gualou Xiebai Baijiu Decoction(GXB)groups(n=9 mice per group).Except for the Control group,the remaining mice were fed a high-fat,high-cholesterol diet for 16 weeks to establish the model,followed by 4 weeks of gavage intervention according to their respective group.Plaque and vascular morphology were observed by Oil red O and hematoxylin-eosin(HE)staining.Serum triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),and serum/intestinal fluid nitrate(NO3-),nitrite(NO2-),and nitric oxide(NO)levels were measured.Macro-transcriptomic sequencing was applied to analyze GM structure and function.NR and nitrite reductase(NiR)activities in serum and intestinal fluid were assessed to validate the result of the macro-transcriptomic functional analysis.Results The pathological progression of AS was significantly improved in the GXB group compared with the Model group,while TG,TC,and LDL-C levels exhibited downward trends and HDL-C showed an opposite upward tendency.NO3-,NO2-,and NO levels in serum and intestinal fluids were significantly elevated.GXB reduced the richness of the GM in AS mice and improved its evenness.It also significantly decreased the abundance of Firmicutes and increased the abundance of anaerobic bacteria including Bacteroidetes and Verrucomicrobia.Functional analysis revealed that GXB promoted the activity of NR,activating the NO3--NO2--NO pathway,which increased endogenous NO levels.The GXB group also showed higher levels of serum/intestinal fluid NR and NiR compared with the Model group,consistent with the functional analysis.Conclusions GXB may improve the structure and function of the GM,increase the abundance of anaerobic gut bacteria,promote NR activity,activate the NO3--NO2--NO pathway,and elevate endogenous NO levels,thereby improving dyslipidemia and alleviating pathological AS damage.
