"One Health"监测中IncP6型质粒基因组分析与全球质粒序列比较OA
IncP6 plasmid genomic analysis in"One Health"surveillance and comparison of global plasmid sequences
目的 基于"One Health"视角,探究携带编码肺炎克雷伯菌碳青霉烯酶(KPC)的blaKPC-2基因的IncP6型质粒的全球传播规律及基因组特征.方法 基于Plasmids in Pathogens Database(PIPdb)分析IncP6质粒的全球分布特征.从中国河南省采集人群、动物、环境及食品来源样本,筛选碳青霉烯耐药菌株,对耐药菌株进行质粒型别鉴定,对筛查到的含IncP6型质粒的碳青霉烯耐药菌株进行质谱初步鉴定及药敏试验,并联合二代和三代测序技术获取菌染色体和质粒基因组序列,并分析携带blaKPC-2基因的IncP6质粒序列与全球公共数据库中相似质粒序列的遗传关联性.结果 IncP6质粒呈现跨洲际传播,可被17个属39个种的细菌携带,存在于动物、人群、食品及环境样本.携带blaKPC-2基因的IncP6型质粒同样具有多宿主适应性、广地理分布及跨生态位存活特性.从河南环境来源分离的携带含blaKPC-2基因的IncP6型质粒来自一株解鸟氨酸拉乌尔菌,该菌呈多重耐药表型、且携带多种耐药基因.本研究检出的IncP6质粒与公共数据库中质粒存在一定的遗传距离,携带广泛传播的ISKpn6/blaKPC-2/截短型blaTEM-1/ISKpn27核心序列及新型可移动元件IS15DIV/msrE/mphE/IS15DIV.结论 IncP6质粒在全球已广泛存在,其携带blaKPC-2基因的IncP6型质粒具有遗传结构上的稳定性和传播力,已在动物-人群-环境间转移传播,基于"One Health"视角下监测携带blaKPC-2基因的IncP6型质粒的基因组特征和全球传播具有重要意义.
This study was aimed at exploring the global dissemination and genomic characteristics of IncP6-type plasmids carry-ing the Klebsiella pneumoniae carbapenemase(KPC)-encoding blaKPC-2 from a One Health perspective.We analyzed global distribu-tion features of IncP6 plasmids by using the Plasmids in Pathogens Database(PIPdb).Human-,animal-,environmental-,and food-derived samples were collected from Henan Province,China,under the One Health framework.We subsequently screened for carbapenem-resistant strains,performed plasmid incompatibility group typing on the resistant strains,and conducted preliminary identification and antimicrobial susceptibility testing on the screened carbapenem-resistant strains bearing IncP6-type plasmids.Se-quences of chromosomal and plasmid genomes were obtained through combined second-and third-generation sequencing technolo-gies.The genetic correlation between the IncP6 plasmid sequences carrying the blaKPC-2 gene and similar plasmid sequences in global public databases was analyzed.IncP6 plasmids demonstrated pandemic spread;were carried by bacteria from 39 species across 17 gen-era;and were detected in animal,human,food,and environmental samples.The blaKPC-2-carrying IncP6 plasmids exhibited broad host adaptability,extensive geographical distribution,and cross-niche persistence.An environmental isolate of Raoultella ornithino-lytica from Henan carried an IncP6 plasmid bearing blaKPC-2.This strain displayed a multidrug-resistant phenotype with multiple resis-tance genes.The identified IncP6 plasmid showed a certain genetic distance from related plasmids in public databases,and carried both the epidemic core resistance structure(ISKpn6/blaKPC-2/ΔblaTEM-1/ISKpn27)and a novel mobile genetic element(IS15DIV/msrE/mphE/IS15DIV).The IncP6 plasmid is widely present globally.The blaKPC-2-carrying IncP6 plasmid demonstrated stability and trans-missibility,and has been transferred across animal-human-environmental compartments.Future monitoring of the molecular charac-teristics and global spread of the blaKPC-2-carrying IncP6 plasmid from a One Health perspective is therefore important.
