云芝多糖对急性肺损伤小鼠的保护作用及对肠道菌群的影响OA
Coriolus versicolor polysaccharide's protective effect on mice with acute lung injury and its influence on gut microbiota
目的:探究云芝多糖对急性肺损伤(ALI)小鼠的保护作用及对肠道菌群的影响.方法:将24只小鼠随机分成三组,FC 组(空白组)、FM 组(LPS 模型组)、FY 组(云芝多糖预防组).FY 组的小鼠每天灌胃 200 mg/kg 云芝多糖,FC 组和 FM 组的小鼠每天灌胃等量的生理盐水,连续灌胃处理 15 d;24 h 后对 FM 组和 FY 组的小鼠进行气管滴注 13 mg/kg LPS,建立 ALI 模型.造模 12 h 后,对小鼠采样.检测小鼠血清中IL-6、IL-10、IL-β、TNF-α、T-AOC、SOD、GSH-Px 和MDA 的水平;采用 HE 染色观察肺组织病理形态学变化;采用测序技术分析小鼠肠道中菌群变化.结果:与 FM 组比较,FY 组小鼠肺组织的炎症细胞浸润均减少,肺泡隔增厚、肺泡水肿和毛细血管充血等情况均有所改善;血液中 IL-6、IL-1β、TNF-α 水平均显著降低,抗氧化能力显著提高.肠道菌群分析发现云芝多糖调节了小鼠肠道菌群的相对丰度,增加乳酸杆菌、放线菌、双歧杆菌等益生菌的相对丰度,并降低了普雷沃菌等有害菌的相对丰度,维持了肠道稳态.结论:云芝多糖可以通过减轻炎症反应、增强抗氧化能力和调节小鼠肠道菌群来发挥抗 ALI 的作用.
AIM:To investigate the protective ef-fect of Coriolus versicolor polysaccharide on acute lung injury(ALI)mice and its influence on the in-testinal microbiota.METHODS:Twenty-four mice were randomly divided into three groups:FC group(control group),FM group(LPS induced group),and FY group(Cordyceps polysaccharide prevention group).Mice in the FY group were given 200 mg/kg Coriolus versicolor polysaccharide by gavage daily,while mice in the FC group and FM group were giv-en the same volume of normal saline by gavage dai-ly for 15 days.24 hours later,LPS(13 mg/kg)was in-tratracheally instilled into mice in the FM group and FY group to establish the ALI model.12 hours after the model was established,samples were collected from the mice.The levels of IL-6,IL-10,IL-1β,TNF-α,T-AOC,SOD,GSH-Px,and MDA in the serum of the mice were detected;the pathological morphology changes of the lung tissue were observed via HE staining;and the changes in the intestinal microbio-ta of the mice were analyzed by sequencing tech-nology.RESULTS:Compared with the FM group,the inflammatory cell infiltration in the lung tissue of mice in the FY group was reduced,and the thicken-ing of the alveolar septum,pulmonary edema,and congestion of capillaries were improved;the levels of IL-6,IL-1β,and TNF-α in the blood were signifi-cantly decreased,and the antioxidant capacity was significantly increased.The analysis of the intestinal microbiota revealed that Coriolus versicolor polysaccharide regulated the relative abundance of the intestinal microbiota in mice,increasing the rel-ative abundance of probiotics such as Lactobacillus,Actinomyces,and Bifidobacterium,and reducing the relative abundance of harmful bacteria such as Prevotella,maintaining intestinal homeostasis.CONCLUSION:Coriolus versicolor polysaccharide can exert anti-ALI effects by reducing inflammatory responses,enhancing antioxidant capacity,and reg-ulating the intestinal microbiota of mice.
