目的 探讨艾司氯胺酮(ESK)对卵巢癌细胞增殖的影响及作用机制.方法 体外培养卵巢癌SKOV3细胞,用不同浓度(0.5 mmol/L、1.0 mmol/L、2.0 mmol/L)的ESK处理SKOV3细胞,同时设置对照组(0 mmol/L ESK).采用CCK-8法和平板克隆法分别检测细胞增殖和克隆形成;采用流式细胞术检测细胞凋亡和细胞周期分布;采用Western blot检测细胞中Wnt/β-catenin通路相关蛋白Wnt3a和β-catenin蛋白,凋亡相关蛋白B细胞淋巴瘤-2(Bcl-2)、胱天蛋白酶-3(Caspase-3)和Bcl-2相关X蛋白(Bax)蛋白,增殖相关蛋白Ki67和cyclin D1蛋白的表达.结果 与对照组比较,不同浓度ESK处理组的SKOV3细胞增殖抑制率、凋亡率和细胞周期G0/G1期占比及Bax、Caspase-3蛋白表达均明显升高(P<0.05),克隆形成数目、细胞周期G2/M期占比及Wnt3a、β-catenin、Ki67、cyclin D1、Bcl-2蛋白表达均明显降低(P<0.05),并均呈浓度依赖性(P<0.05).结论 ESK能够抑制卵巢癌细胞增殖,其作用可能是通过调控Wnt/β-catenin信号通路实现的.

Objective To investigate the effects of esketamine(ESK)on the proliferation of ovarian cancer cells and its mechanism of action.Methods Ovarian cancer SKOV3 cells were cultured in vitro and treated with different concentrations(0.5 mmol/L,1.0 mmol/L,2.0 mmol/L)of ESK,while the control group(0 mmol/L ESK)was established simultaneously.Cell proliferation and colony formation were detected by CCK-8 assay and colony formation assay,respectively.Flow cytometry was used to detect cell apoptosis and cell cycle distribution.Western blot was used to detect the expression levels of Wnt/β-catenin pathway-related proteins(Wnt3a and β-catenin),apoptosis-related proteins[B-cell lymphoma-2(Bcl-2),Caspase-3,and Bcl-2-associated X protein(Bax)],and proliferation-related proteins(Ki67 and cyclin D1).Results Compared with the control group,ESK treatment at different concentrations significantly increased the proliferation inhibition rate of SKOV3 cells,apoptosis rate,percentage of cells in the G0/G1 phase,as well as the expression of Bax and Caspase-3 proteins(P<0.05).Meanwhile,the number of colonies,percentage of cells in the G2/M phase,and the expression levels of Wnt3a,β-catenin,Ki67,cyclin D1,and Bcl-2 proteins were significantly decreased(P<0.05),showing a concentration-dependent effect(P<0.05).Conclusion ESK can inhibit the proliferation of ovarian cancer cells,and its effect may be achieved by regulating the Wnt/β-catenin signaling pathway.
