山东省部分健康猪和病死猪源沙门菌血清型、耐药基因及基因型分析OA
Analysis of Serotypes,Antibiotic Resistance Genes and Genotypes of Salmonella Isolated from Healthy and Dead Pigs in Shandong Province
为了解山东省健康猪和病死猪中的沙门菌血清型、耐药基因和基因型分布特征,对从养殖场、屠宰场和无害化处理场收集的375份猪肛拭子和淋巴结样品进行沙门菌分离鉴定,对分离株采用K-B药敏纸片法检测产超广谱β-内酰胺酶(ESBLs)表型,用PCR法检测血清型以及15种耐药基因和多序列位点分型(MLST)分布,通过脉冲场凝胶电泳(PFGE)分析鼠伤寒沙门菌(S.typhimurium)及单相变种(S.1,4,[5],12:i:-)菌株间的遗传关系.结果显示:共分离获得48株沙门菌(12.80%),其中病死猪(24/135,17.78%)、淋巴结样品(44/255,17.25%)的分离率最高.共鉴定出8种血清型,其中鼠伤寒沙门菌单相变种和里森沙门菌(S.rissen)在健康猪和病死猪中都是优势血清型,肠炎沙门菌(S.enteritidis)只在病死猪中被分离到.共鉴定出9种ST型,其中ST11(10/24,41.67%)、ST34(5/24,20.83%)和ST469(5/24,20.83%)是病死猪源沙门菌的优势ST型,而ST34(6/24,25.00%)、ST469(6/24,25.00%)和ST19(5/24,20.83%)是健康猪源的优势ST型.此外,在18株鼠伤寒沙门菌及单相变种中共鉴定出13种PFGE谱型,病死猪和健康猪源沙门菌的谱型不同.耐药基因qnrA、blaSHV、blaTEM-1、tetA检出率均高于60%,其中blaSHV检出率病死猪源沙门菌高于健康猪源,su1 Ⅱ、tetA、aacC3检出率则健康猪源沙门菌高于病死猪源.7株产ESBLs沙门菌中,健康猪和病死猪源菌株分别有3株和4株,且全部携带β-内酰胺和喹诺酮类耐药基因.结果表明:山东省病死猪和健康猪源沙门菌的优势血清型、分子型都有差异,且呈现多态性,耐药基因谱复杂多样.建议加强对山东省猪源沙门菌及其耐药情况的监测,为有效的沙门菌控制净化以及指导临床合理用药提供更多依据.
In order to investigate the distribution of serotypes,antibiotic resistance genes and genotypes of Salmonella isolated from healthy and dead pigs in Shandong Province,375 swine anal swab and lymph node samples were collected from farms,slaughterhouses and rendering plants to isolate and identify Salmonella.The isolates were detected for ESBLs phenotype by K-B disk diffusion susceptibility test,for serotypes,15 antibiotic resistant genes and MLST distribution by PCR,and for identification of genetic relationships between S.typhimurium and monophasic variant(S.1,4,[5],12:i:-)by PFGE.The results showed that 48 Salmonella strains(12.80%)were isolated,most from dead pigs(24/135,17.78%)and lymph node samples(44/255,17.25%).8 serotypes were identified,dominated by monophasic variant of S.typhimurium and S.rissen in both healthy and dead pigs,but S.enteritidis was only isolated from the latter.A total of 9 ST types were identified,dominated by ST 11(10/24,41.67%),ST34(5/24,20.83%)and ST469(5/24,20.83%)in Salmonella isolates from dead pigs,but ST34(6/24,25.00%),ST469(6/24,25.00%)and ST 19(5/24,20.83%)in those from healthy pigs.Furthermore,13 PFGE spectrotypes were identified from 18 isolates of S.typhimurium and its monophasic variant,and different between those derived from dead and healthy pigs.The detection rates of resistance genes qnrA,blaSHV,blaTEM-1 and tetA were all over 60%,among which that of blaSHV was higher in isolates from dead pigs than in those from healthy pigs,while those of sul Ⅱ,tetA and aacC3 were higher in isolates from healthy pigs than in those from dead pigs.There were 7 strains of ESBLs-producing Salmonella carrying both β-lactam and quinolone resistance genes,3 and 4 were from healthy and dead pigs,respectively.In conclusion,the predominant serotypes and molecular types of Salmonella isolates from healthy and dead pigs were different,characterized by polymorphism as well as complex and diverse antibiotic resistance gene spectrum.Therefore,porcine Salmonella and its antimicrobial resistance should be further monitored in Shandong Province,with a view to providing more evidence for effective control and eradication of Salmonella,as well as for guiding prudent use of drugs in practice.
