为建立高灵敏度、高特异性且能够绝对定量的牛传染性鼻气管炎病毒(IBRV)检测方法,该研究基于IBRV的gB基因设计特异性引物、探针,建立了IBRV微滴式数字PCR检测方法,对其退火温度、引物/探针浓度等反应条件进行优化,并对该方法的特异性、灵敏性以及符合率进行验证.结果表明,该方法特异性良好,能特异性检测牛传染性鼻气管炎病毒,与口蹄疫病毒、牛病毒性腹泻病毒、牛呼吸道合胞体病毒、牛3型副流感病毒等均无交叉反应;灵敏度高,最低检测限可达0.94 copies/μL;符合率高,与qPCR标准检测方法符合率100%.以上结果表明,建立的IBRV ddPCR检测方法特异性强、灵敏度高、符合率高的优势,可满足IBRV精准定量、早期诊断、疫苗抗原定量、进出口检疫及流行病学监测的需求,为牛传染性鼻气管炎的精准检测提供技术支撑.

To establish a detection method for IBRV with high sensitivity,high specificity,and absolute quantification capability,this study designed specific primers and probes targeting the gB gene of IBRV,thereby developing an IBRV droplet digital PCR(ddPCR)assay.Critical reaction parameters including annealing temperature and primer/probe concentrations were systematically optimized,followed by validation of the method's specificity,sensitivity,and concordance.The results demonstrated that:the assay exhibited excellent specificity,specifically detecting IBRV without any cross-reactivity with FMDV,BVDV,BRSV,BPIV 3.It showed high sensitivity,with a minimum detection limit as low as 0.94 copies/μL.It achieved a 100%concordance rate with the standard qPCR detection method,indicating high consistency.These findings confirm that the established IBRV ddPCR assay,characterized by strong specificity,high sensitivity,and excellent concordance,can meet the requirements for precise quantification of IBRV,early diagnosis,vaccine antigen quantification,import-export quarantine,and epidemiological surveillance.It thus provides technical support for the accurate detection of Infectious Bovine Rhinotracheitis.
