为建立犬瘟热病毒临床快速检测方法,该研究针对犬瘟热病毒N基因保守序列设计RT-LAMP内引物、外引物和环引物,通过反应条件优化、敏感性试验、特异性试验和临床样品检测,建立了CDV RT-LAMP检测方法.结果显示,反应最适温度为63℃,敏感性最低可检测到质粒浓度5.68×101 copies/μL,结合临床样品验证试验,所建CDV RT-LAMP检测方法特异性高,敏感性高于国标RT-PCR检测方法.可应用于犬瘟热临床快速诊断,为下一步技术推广和犬瘟热防控提供了技术支撑.

In order to establish a rapid clinical detection method for canine distemper virus,RT-LAMP internal primers,external primers and loop primer were designed for the study based on the conserved sequences of N gene of canine distemper virus.RT-LAMP assay was established by optimization of reaction conditions,sensitivity test,specificity test and clinical samples test.The results showed that the optimal reaction temperature was 63℃,and the lowest detectable plasmid concentration was 5.68×101 copies/μL.Combined with the clinical sample verification test,the established CDV RT-LAMP detection method had high specificity and higher sensitivity than the national standard RT-PCR detection method.It can be applied to the rapid clinical diagnosis of canine distemper and provides technical support for the next step of technology promotion and canine distemper prevention and control.
