该试验旨在研究桑黄硒多糖(SHXDT)对甲基硫氧嘧啶(MTU)诱导小鼠肝损伤模型的保护作用及潜在机制.试验将小鼠随机分为5组,即正常对照组(NC)、模型组、SHXDT低、中、高剂量组.模型组与各给药组小鼠均腹腔注射MTU以造肝损伤模型,正常对照组注射等量生理盐水.连续处理3周后,检测小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)总胆红素(TBIL)水平,评估肝脏损伤程度;测定肝脏组织中丙二醛(MDA)含量、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,衡量氧化应激水平;通过苏木精—伊红(HE)染色观察肝脏组织病理学变化.结果显示,与模型组相比,SHXDT各剂量组小鼠血清ALT、AST、TBIL水平显著降低;SOD、GSH-Px活性也显著降低;肝脏病理损伤得到改善.结论:SHXDT对MTU诱导的小鼠肝损伤具有显著保护作用.

The aim of this experiment is to investigate the protective effect and potential mechanism of Sanghuang selenium polysaccharide(SHXDT)on a mouse liver injury model induced by methylthiouracil(MTU).The experiment randomly divided the mice into 5 groups,namely the normal control group(NC),the model group,and the low,medium,and high dose groups(SHXDT).The model group and each treatment group of mice were intraperitoneally injected with MTU to create a liver injury model,while the normal control group was injected with an equal amount of physiological saline.After 3 weeks of continuous treatment,the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),total bilirubin(TBIL)in the serum of mice were measured to evaluate the degree of liver damage;Measure the content of malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)activity in liver tissue to assess oxidative stress levels;Observe the pathological changes of liver tissue through hematoxylin eosin(HE)staining.The results showed that compared with the model group,the serum ALT,AST,and TBIL levels of mice in each dose group of SHXDT were significantly reduced;The activities of SOD and GSH-Px also significantly decreased;The pathological damage to the liver has been improved.Conclusion:SHXDT has a significant protective effect on MTU induced liver injury in mice.
