不同固定与染色条件对公牛精液抹片效果的影响研究OA
Effects of Different Fixation and Staining Conditions on Bull Semen Smear Quality
该研究旨在优化精子畸形率检测中的精液涂片制备方案,以提高检测的准确性与效率.通过比较抹片固定时间(0.5、1、2、3 min)、染色时间(1、2、3、5 min)、精液体积(5、8、10 μL)及染色液种类(龙胆紫、结晶紫),评估其对抹片背景深浅与精子着色效果的影响.结果表明,5 μL精液所制抹片效果优于8 μL和10 μL.在该体积下,龙胆紫染色效果优于结晶紫.固定3 min结合染色3 min或5 min会导致背景偏深,而固定0.5 min则精子着色不佳.综合效果与时间效率,固定1~2 min、染色1~2 min为较优参数区间.综上可知,采用5 μL精液、95%乙醇固定1 min、0.1%龙胆紫染色1 min的涂片方案,可制备背景清晰、精子形态明显的抹片,符合国家标准要求,适用于精子畸形率鉴定,具有良好的推广应用价值.
This study aimed to optimize semen smear preparation methods for sperm morphological abnormality assessment,thereby improving the accuracy and efficiency of detection.By comparing fixation times(0.5,1,2,and 3 min),staining times(1,2,3,and 5 min),semen volumes(5,8,and 10 μL),and staining reagents(gentian violet and crystal violet),the effects of these parameters on smear background intensity and sperm staining quality were assessed.The results showed that smears prepared with 5 μL semen yielded better outcomes than those prepared with 8 μL and 10 μL.At this volume,gentian violet produced superior staining effects compared with crystal violet.Fixation for 3 min combined with staining for 3 or 5 min resulted in darker backgrounds,while fixation for 0.5 min led to insufficient staining of spermatozoa.Considering both staining quality and time efficiency,fixation for 1~2 min and staining for 1~2 min were identified as the optimal parameter range.Ultimately,use 5 μL of semen,fixation for 1 min with 95%ethanol and staining with 0.1%gentian violet for 1 min was determined to be the optimal combination,producing smears with clear backgrounds and well-defined sperm morphology.This protocol meets the requirements for sperm abnormality assessment and has considerable potential for practical application.
