为探究绵羊骨形态发生蛋白15(BMP15)基因FecXGr突变对其编码蛋白功能的潜在影响,该研究采用生物信息学方法,对比分析突变前后蛋白理化性质、二级/三级结构、翻译后修饰位点及跨物种保守性.结果显示,FecXGr突变未改变BMP15蛋白碱性属性、亲水性、分泌特性及3个保守糖基化位点,仅使蛋白分子量微升至44.91 kDa、总原子数增至6 347;磷酸化苏氨酸位点减少1个;二级结构中α螺旋与延伸链占比下降、无规则卷曲占比升高,β折叠与三级结构稳定.跨物种保守性分析表明,绵羊BMP15核苷酸及其编码的蛋白序列与反刍动物(山羊、牛)一致性最高,与人类最低,显示该基因在反刍动物中高度保守.该研究为解析FecXGr突变调控绵羊繁殖性能机制及分子育种提供理论参考.

To investigate the potential impact of the FecXGr mutation in the sheep bone morphogenetic protein 15(BMP15)gene on the function of its encoded protein,this study employed bioinformatics methods to compare the physicochemical properties,secondary/tertiary structures,post-translational modification sites,and cross-species conservation of the protein before and after the mutation.Results indicate that the FecXGr mutation did not alter the BMP15 protein's basicity,hydrophilicity,secretory properties,or three conserved glycosylation sites.It only slightly increased the protein molecular weight to 44.91 kDa and total atom count to 6 347;reduced phosphorylation sites by one;The proportion of α-helices and extended chains in the secondary structure decreased,while the proportion of disordered regions increased.β-folds and the tertiary structure remained stable.Cross-species conservation analysis indicated that sheep BMP15 nucleotide sequences and their encoded protein sequences showed the highest consistency with ruminants(goats,cattle)and the lowest with humans,demonstrating high conservation of this gene in ruminants.This study provides theoretical insights for elucidating the mechanism by which the FecXGr mutation regulates sheep reproductive performance and for molecular breeding.
