大黄素通过miR-375/KLF5轴减轻变应性鼻炎炎症OA
Effects of emodin on allergic rhinitis inflammation through the miR-375/KLF5 axis
探究大黄素(EM)对变应性鼻炎(AR)小鼠炎症反应的影响.采用酶联免疫吸附法、苏木素-伊红和过碘酸雪夫氏染色、定量实时聚合酶链式反应、蛋白免疫印迹法检测血清中卵清蛋白(OVA)、特异性免疫球蛋白E(IgE)和组胺的水平、鼻腔灌洗液(NALF)中炎性细胞数量及炎性细胞因子水平、鼻黏膜组织的病理变化、鼻黏膜组织中miR-375和Kruppel样因子5(KLF5)mRNA水平和KLF5蛋白表达水平.结果发现经EM处理小鼠血清OVA特异性IgE和组胺水平,NALF中嗜酸性粒细胞、嗜中性粒细胞、巨噬细胞和淋巴细胞数量,IL-4、IL-5和IL-13的质量浓度以及鼻黏膜组织中miR-375表达均降低.此外,鼻黏膜组织中杯状细胞增生减少,炎症细胞浸润得到缓解,而KLF5 mRNA和KLF5蛋白表达水平升高,(P<0.05).结果表明EM可以通过下调miR-375,促进KLF5表达来抑制AR小鼠的炎症反应.
This study aimed to investigate the effects of emodin(EM)on inflammation in mice with allergic rhinitis(AR).Serum ovalbumin(OVA)-specific immunoglobulin E(IgE)and histamine levels,inflammatory cell counts,and cytokine levels in nasal lavage fluid(NALF)were measured.Pathological changes in nasal mucosal tissue,along with miR-375 and KLF5 mRNA levels,and KLF5 protein expression were assessed using enzyme-linked immunosorbent assay,hematoxylin-eosin and periodic acid-Schiff staining,quantitative real-time polymerase chain reaction,and protein immunoblotting.The results showed that EM treatment reduced serum OVA-specific IgE and histamine levels,the counts of eosinophils,neutrophils,macrophages,and lymphocytes in NALF,as well as the mass concentrations of IL-4,IL-5,and IL-13,along with miR-375 expression in nasal mucosal tissue.Additionally,the hyperplasia of goblet cells in the nasal mucosa tissue was reduced,the infiltration of inflammatory cells was alleviated,while the expression of KLF5 mRNA and KLF5 protein increased,(P<0.05).These findings indicate that EM can inhibit the inflammatory response in AR mice by downregulating miR-375 and promoting KLF5 expression.
