不同种源大白杜鹃种子表型性状和发芽特性分析及其综合评价OA

Analysis and comprehensive evaluation on seed phenotypic traits and germination characteristics of Rhododendron decorum from different provenances

[目的]探明不同种源大白杜鹃种子表型性状及发芽特性,为大白杜鹃的良种选育提供支撑.[方法]以贵州6个种源(雷公山自然保护区、六盘水市钟山区、百里杜鹃管理区、威宁县、赫章县和麻江县)的大白杜鹃种子为研究对象,采用相关分析、聚类分析、主成分分析和综合评价等方法,分析不同种源种子的表型性状(种子长、种子宽、种子厚、种翅长、种翅宽、大小指数、长宽比、扁化率以及千粒重)与发芽性状(发芽势、发芽率、发芽指数、活力指数、胚根长、胚芽长)的变异特征及遗传特性.[结果]不同种源大白杜鹃种子表型性状间除长宽比外均存在显著差异(P<0.05);表型性状变异系数和遗传力分别为9.64%~43.56%和0.522~0.978.种子长、种子宽、种翅长、种翅宽以雷公山、麻江较大,种子厚以威宁、赫章较大,大小指数以赫章最大,扁化率以百里杜鹃最大,千粒重以麻江最大.发芽性状除发芽率、胚芽长在种源间差异达显著水平外,其他性状间均无显著差异;发芽性状变异系数和遗传力分别为8.75%~30.01%和0.34~0.84;发芽率以百里杜鹃最低(32.3%),胚芽长以雷公山最长(2.15 mm).相关分析表明,发芽率、发芽指数与种子厚及大小指数呈显著或极显著正相关,与扁化率呈显著或极显著负相关;纬度与发芽率、发芽指数呈显著负相关,海拔与活力指数呈显著正相关,无霜期与活力指数、胚根长呈极显著或显著负相关;经度、年均气温、年均降水量及无霜期与种子长、种子宽、种翅长、种翅宽间均存在显著或极显著正相关.6个种源大白杜鹃聚为3个类群,其中第一类群为雷公山、麻江,第二类群为钟山、威宁和赫章,第三类群为百里杜鹃.不同种源大白杜鹃综合得分为雷公山>麻江>赫章>威宁>钟山>百里杜鹃.[结论]不同种源大白杜鹃种子的表型性状存在显著差异,并具有高遗传变异和高遗传力的特点,雷公山种源可作为遗传育种及种子繁殖的种源进一步研究,百里杜鹃需要进行就地保护和迁地保护,以防止种群衰退和遗传资源的流失.

[Objective]The phenotypic traits and germination characteristics of Rhododendron decorum seeds from different provenances were explored to provide a scientific support for breeding of improved R.decorum varieties.[Method]The variation and hereditary characteristics in phenotypic traits(seed length,seed width,seed thickness,samara length,samara width,seed size index,length-width ratio,flatness rate and 1000-seed weight)and germination traits(germination potential,germination rate,germination index,vigor index,radicle length,plumule length)of R.decorum seeds from six provenances in Guizhou province(Leigongshan National Nature Reserve,Zhongshan district in Liupan-shui city,Baili Dujuan Management District,Weining county,Hezhang county and Majiang County)were analyzed by correlation analysis,cluster analysis,principal component analysis,and comprehensive evaluation methods.[Result]There were significant differences(P<0.05)in seed phenotypic traits among different provenances except for length to width ratio.The variable coefficient and heritability of seed pheno-typic traits were 9.64%-43.56%and 0.522-0.978,respectively.The seed length,seed width,samara length,and samara width of R.de-corum seeds from Leigongshan and Majiang provenances were higher.The maximum seed thickness,seed size index,flatness rate and 1000-seed weight of R.decorum seeds from Weining and Hezhang provenances,Hezhang provenance,Baili Dujuan provenance,and Majiang prove-nance,respectively.There were no significant differences in seed germination traits among 6 provenances except for seed germination rate and plumule length.The variable coefficient and heritability of seed germination traits were 8.75%-30.01%and 0.34-0.84,respectively.The lowest seed germination rate(32.3%)and highest plumule length(2.15 mm)of R.decorum seeds were from Baili Dujuan provenance and Leigongshan provenance,separately.The seed germination rate and germination index were positively related to seed thickness and seed size index significantly or very significantly,and negatively related to seed flatness rate significantly or very significantly.The latitude was negative-ly related to seed germination rate and germination index significantly.The altitude was positively and significantly related to vigor index.The frost-free period was negatively related to vigor index and radicle length significantly or very significantly.The longitude,annual mean temper-ature,annual mean precipitation and frost-free period all were positively correlated with seed length,seed width,samara length and width sig-nificantly or very significantly.Six R.decorum provenances were clustered into three groups of Group Ⅰ(Leigongshan and Majiang),Group Ⅱ(Zhongshan,Weining,and Hezhang)and Group Ⅲ(Baili Dujuan).The comprehensive score of six R.decorum provenances was Leigongs-han>Majiang>Hezhang>Weining>Zhongshan>Baili Dujuan.[Conclusion]There are significant differences in phenotypic traits of R.de-corum seeds among six provenances,and the phenotypic traits of R.decorum seeds have the higher heritable variation and heritability.The R.decorum seeds from Leigongshan provenance can be used as a potential source for genetic breeding and seed propagation.The R.decorum seeds from Baili Dujuan provenance should be conserved in situ and ex situ to prevent population decline and genetic resources loss.
