基于SSR标记的63份芥菜型油菜种质资源的遗传多样性和群体结构分析OA
Genetic diversity and population structure analysis of 63 mustard type rapeseed(Brassica juncea)germplasm resouces based on SSR markers
[目的]探究63份芥菜型油菜种质资源间的遗传多样性和群体结构,为挖掘优异等位变异、提高芥菜型油菜育种效率提供理论依据,加快育种进程.[方法]以63份来源不同的芥菜型油菜资源为研究对象,运用SSR分子标记技术对其进行遗传差异分析、聚类分析、群体遗传结构分析及主成分分析.[结果]利用筛选出的均匀分布于芥菜型油菜18条染色体上的54个条带清晰的SSR标记,对63份芥菜型油菜种质资源进行遗传多样性分析,结果共检测到204个等位变异,平均每对引物检测到3.7个等位基因数和1.97个有效等位基因数,群体平均Shannon's遗传多样性指数为0.68,平均Nei's基因多样性指数为0.49,观测杂合度(Ho)平均值为0.58.PIC值为0.28~0.38,平均值为0.36,均为中高多态性引物(PIC>0.25).综上选取的54个SSR标记具有中高多态性,能较好的检测参试材料的遗传多样性,且63份芥菜型油菜资源遗传信息丰富,拥有均匀且丰富的等位基因.另外,利用SSR标记分析结果进行聚类分析和遗传群体结构分析,UPGMA法将63份供试材料聚类成两大类群,类群Ⅰ包含9份材料,类群Ⅱ包含54份材料.利用Structure软件对供试材料进行群体结构分析,当K=2时ΔK出现最大值,由此判断可将63份芥菜型油菜种质资源分为2个组群,这一结果与利用二维主坐标系进行主成分分析的结果相似.组群Ⅰ中Q值大于0.6的有48份种质资源,占所有供试材料的76.19%,Q值大于0.9的有29份种质资源,占所有供试材料的46.03%.综上所述,63份芥菜型油菜种质资源的遗传背景呈现多样性,群体结构较为丰富.[结论]筛选出条带清晰、具有中高多态性、能较好的检测参试材料遗传差异的SSR标记54对,利用54对SSR标记检测分析发现,63份芥菜型油菜种质资源遗传信息丰富,具有较高的遗传差异.
[Objective]The present paper aimed to investigate the genetic diversity and population structure among 63 Brassica juncea germ-plasm resources,providing a theoretical basis for mining superior allelic variations,improving breeding efficiency,and accelerating the breed-ing process.[Method]63 B.juncea accessions from diverse origins were used as research materials,and genetic differentiation analysis,cluster analysis and population genetic structure analysis were conducted employing SSR molecular marker technology.[Result]54 SSR markers with good polymorphism and clear bands,uniformly distributed across the 18 chromosomes of B.juncea,were selected for genetic diversity analy-sis.A total of 204 allelic variations were detected,with an average of 3.7 alleles and 1.97 effective alleles per primer pair.The average Shan-non's genetic diversity index was 0.68,the average Nei's gene diversity index was 0.49,and the average observed heterozygosity(Ho)was 0.58.The PIC values ranged from 0.28 to 0.38,with an average of 0.36,indicating that all primers were moderately to highly polymorphic(PIC>0.25).In summary,the 54 SSR markers exhibited moderate to high polymorphism and effectively detected the genetic diversity of the tested materials.Additionally,the 63 B.juncea accessions displayed rich genetic information,with uniform and abundant allelic variations.Furthermore,cluster analysis and population genetic structure analysis were performed based on the SSR marker results.The UPGMA method classified the 63 accessions into two major groups:Group Ⅰ comprised 9 accessions,while Group Ⅱ included 54 accessions.Population struc-ture analysis using the structure software revealed that the maximum ΔK occurred at K=2,indicating that the 63 B.juncea accessions could be divided into two subpopulations.The result was consistent with the two-dimensional principal coordinate analysis(PCoA).In Subpopula-tion Ⅰ,48 accessions(76.19%of the total)had Q-values greater than 0.6,and 29 accessions(46.03%of the total)hadQ-values greater than 0.9.These results demonstrated that the 63 B.juncea accessions exhibited diverse genetic backgrounds and a rich population structure.[Conclusion]54 SSR markers with clear bands,moderate to high polymorphism,and effective detection of genetic differences are selected.A-nalysis using these markers reveals that the 63 B.juncea accessions possess abundant genetic information and high genetic diversity.
