毛荚苜蓿DIR基因家族成员的鉴定及表达模式分析OA
Identification and expression pattern analysis of members of DIR gene family in Medicago edgeworthii
[目的]鉴定毛荚苜蓿DIR(Dirigent)基因家族成员,分析MeDIR基因在多种非生物胁迫中的表达模式,为阐明MeDIR基因家族成员的生物学功能奠定基础.[方法]利用生物信息学鉴定毛荚苜蓿基因组中的MeDIR基因家族成员,根据鉴定结果与拟南芥和蒺藜苜蓿构建系统发育树,利用在线网站ExPASy预测MeDIR蛋白理化性质,分析基因家族成员染色体定位、基因结构、顺式作用元件,并预测毛荚苜蓿DIR-a亚家族成员的蛋白三维结构.[结果]在毛荚苜蓿全基因组范围内,共发现32个DIR基因家族的成员,将其依次命名为MeDIR1~MeDIR32,不均匀分布在6条染色体上,部分成员具有内含子和非翻译区.根据系统进化树关系,共分为5个亚族(DIR-a、DIR-b、DIR-c、DIR-d和DIR-e).其中,84%MeDIR蛋白具有较高的稳定性,44%MeDIR蛋白具有疏水性.亚细胞定位显示,有11个MeDIR蛋白定位于叶绿体中,4个MeDIR蛋白定位于内质网中,8个MeDIR蛋白分别定位于胞外和细胞质中,其余少数蛋白分别定位于液泡膜、过氧化氢酶体、细胞骨架和细胞核中,蛋白定位的多样化说明MeDIR蛋白可能发挥多种生物学功能.顺式作用元件分析显示,MeDIR基因启动子区域具有多个生长发育、植物激素和逆境胁迫响应元件.实时荧光定量PCR分析发现,MeDIR基因的表达受干旱、低温和ABA胁迫的差异调控,表明MeDIR基因家族可能参与毛荚苜蓿对非生物胁迫的适应性调控.[结论]MeDIR基因家族成员数量较多、分布广泛,启动子顺式作用元件差异显著,部分MeDIR基因的表达受低温、干旱和ABA处理的诱导,积极响应非生物胁迫.
[Objective]The study aimed to identify the members of the DIR gene family in Medicago edgeworthii,examine the expression pro-files of MeDIR genes in response to diverse abiotic stresses,and lay the foundation for elucidating the biological functions of the MeDIR gene family members.[Method]The MeDIR gene family members were identified through bioinformatics analysis,and a phylogenetic tree was con-structed with their counterparts from Arabidopsis thaliana and M.truncatula.The online tool ExPASy was used to predict the physicochemical properties of the MeDIR family proteins,characterize the chromosomal distribution,gene architecture,and promoter cis-elements of the gene family members,and additionally predict the three-dimensional protein structures of the DIR-a subfamily members in M.edgeworthii.[Re-sult]32 DIR gene family members were identified in the M.edgeworthii genome,designated as MeDIR1 to MeDIR32.They were unevenly dis-tributed across six chromosomes,with some members containing introns and untranslated regions.Based on phylogenetic analysis,they were classified into five subfamilies(DIR-a,DIR-b,DIR-c,DIR-d and DIR-e).84%of the MeDIR proteins exhibited high stability,and 44%were hydrophobic.Subcellular localization predictions indicated that 11 MeDIR proteins were localized in the chloroplast,4 proteins in the en-doplasmic reticulum,and 8 proteins in the extracellular space and cytoplasm,while the remaining were localized in the vacuolar membrane,peroxisomes,cytoskeleton,and nucleus.The diversity of protein localization suggested that MeDIR proteins might perform a variety of biologi-cal functions.Cis-acting element analysis revealed that the promoter regions of MeDIR genes contained multiple elements related to growth and development,plant hormones,and stress responses.Quantitative real-time PCR revealed that the expression of MeDIR genes was differentially regulated under drought,low-temperature,and ABA stress conditions,indicating that the MeDIR gene family might be involved in the adaptive regulation of M.edgeworthii to abiotic stresses.[Conclusion]The MeDIR gene family exhibited a large number of members and wide distribu-tion,with significant differences in promoter cis-acting elements.Some MeDIR genes were induced by low temperature,drought,and ABA treatment,actively responding to abiotic stresses.
