伊犁地区野生鸭茅根腐病调查、病原菌鉴定及其生物学特性OA
Investigation,Pathogen Identification and Biological Characteristics of Root Rot on Wild Dactylis glomerata L.in Yili Region
为明确新疆野生鸭茅(Dactylis glomerata L.)根腐病病原菌种类及其生物学特性.在新疆伊犁昭苏、特克斯、新源县野生鸭茅分布区进行病害调查与采样,病原菌采用组织分离及单孢纯化从病株根部获得;通过致病性测定、形态学观察及ITS、TEF-α、RPB2多基因系统发育分析进行病原鉴定;并以菌丝速率法测定其生物学特性.田间调查发现野生鸭茅根腐病,其自然条件下发病率达20%~72%.经鉴定,引起该病害的病原菌为锐顶镰刀菌(Fusarium acuminatum).该菌菌丝生长最适温度为20℃,最适pH值为7,最适碳源为可溶性淀粉,最适氮源为尿素,在马铃薯胡萝卜培养基(PCA)、燕麦片琼脂培养基(OMA)、马铃薯蔗糖琼脂培养基(PSA)和查氏培养基(Czapek)上生长较快,全黑暗条件利于菌落扩展.
In order to identify the pathogen causing root rot on wild Dactylis glomerata L.in Xinjiang and determine its biological characteristics,disease surveys and sampling were conducted in the distribution areas of wild D.glomerata in Zhaosu,Tekes,and Xinyuan Counties of Ili,Xinjiang.The pathogen was iso-lated from the roots of diseased plants using tissue isolation and single-spore purification.Pathogen identi-fication was performed through pathogenicity tests,morphological observation,and multi-gene phylogenet-ic analysis based on ITS,TEF-α,and RPB2 sequences.Biological characteristics were determined using the mycelial growth rate method.Field surveys revealed that the natural incidence of root rot on wild D.glomerata ranged from 20%to 72%.The pathogen was identified as Fusarium acuminatum.The optimal conditions for mycelial growth were 20℃,pH 7,soluble starch as the carbon source,and urea as the nitrogen source.The fungus grew rapidly on potato-carrot agar(PCA),oat meal agar(OMA),potato-sucrose agar(PSA),and Czapek agar.Complete darkness favored colony expansion.
