Identification and Comprehensive Evaluation of Salt Tolerance of Watermelon Germplasm Resources at Seedling Stage
为明确西瓜种质资源苗期复合盐胁迫下的耐盐性差异,筛选强耐盐优异种质.采用水培法,以100 mmol/L(NaCl∶Na2SO4=1∶1)复合盐溶液处理126份西瓜种质幼苗,测定株高、茎粗、SPAD、地上部鲜质量、地下部鲜质量等13项生长指标,计算耐盐系数;结合相关性分析、主成分分析、隶属函数分析进行耐盐性综合评价.结果表明,复合盐胁迫下,各指标的耐盐系数均存在不同程度变异,变异系数10.11%~77.09%;对各指标进行主成分分析,将13个单项指标转换为3个相互独立的综合指标,累计贡献率达 73.72%,各因子载荷最大的指标为根系总表面积、根系平均直径和株高,相关系数分别为0.937、0.715、0.799,可作为西瓜苗期的耐盐主要评价指标;根据综合D值大小将126份西瓜种质划分为5类,其中强耐盐型种质7份,耐盐型种质32份,中等耐盐型种质50份;盐敏感型种质34份;盐极敏感型材料3份.筛选获得强耐盐种质7份:TS74、TS38、ZS19、KS26、ZS30、KS8、ZS32;在相同试验条件下,根系总表面积、根系平均直径和株高可作为西瓜苗期耐盐性鉴定的参考指标.
To clarify the differences in salt tolerance among watermelon germplasm resources at the seed-ling stage under combined salt stress,and to screen out elite germplasm with strong salt tolerance.A hy-droponic culture method was adopted in the process during which seedlings of 126 watermelon germplasm resources were treated with a 100 mmol/L mixed salt solution(NaCl∶Na2SO4=1∶1).Thirteen growth-related traits,including plant height,stem diameter,SPAD value,shoot fresh mass,and root fresh mass were measured to calculate the salt tolerance coefficient.A comprehensive evaluation of salt tolerance was performed by combining correlation analysis,principal component analysis,and membership function anal-ysis.The results showed that under combined salt stress,the salt tolerance coefficients of all indicators ex-hibited variations to different degrees,with the coefficient of variation ranging from 10.11%to 77.10%.Principal component analysis was performed on the indicators,converting 13 individual indicators into 3 mutually independent comprehensive indicators with a cumulative contribution rate of 73.72%.The in-dicators with the highest factor loadings were total root surface area,average root diameter,and plant height,with correlation coefficients of 0.937,0.715,and 0.799 respectively,which could serve as the main evaluation indicators for salt tolerance of watermelon at the seedling stage.Based on the comprehensive D value,126 watermelon germplasm resources were classified into 5 categories,including 7 highly salt-tolerant germplasms,32 salt-tolerant germplasms,50 moderately salt-tolerant germplasms,34 salt-sensitive germ-plasms,and 3 highly salt-sensitive germplasms.Seven germplasms with strong salt tolerance were screened out,namely TS74,TS38,ZS19,KS26,ZS30,KS8 and ZS32.Under the same experimental condi-tions,total root surface area,average root diameter and plant height can be used as reference indicators for the identification of salt tolerance in watermelon at the seedling stage.
