针对新疆特有药用葡萄品种琐琐葡萄(Vitis vinifera Suosuo),在初代培养、增殖培养、生根培养过程中进行培养基和植物生长调节剂的筛选,为其建立高效的茎段快繁体系.以琐琐葡萄单芽茎段为外植体,初代培养由 MS、WPM、B5、Nitsch、ER培养基中筛选腋芽萌发的最优基础培养基;增殖培养采用二次回归正交旋转组合法,分析6-苄基腺嘌呤(6-BA)、吲哚丁酸(IBA)、赤霉素(GA3)配比对芽分化、增殖及再生的影响;生根培养以1/2 MS为基础培养基,设置不同IBA和活性炭(Ac)浓度,并进行不同时长的暗培养,筛选最优生根配方与暗处理条件.最适琐琐葡萄茎段腋芽萌发的培养基为 MS培养基,并添加2.00 mg/L 6-BA+0.20 mg/L IBA+0.20 mg/L GA3,其腋芽萌发率达90.00%;二次回归正交旋转组合分析显示,影响茎段增殖的植物生长调节剂主次顺序为6-BA>IBA>GA3,最适宜的增殖配方为 MS+1.77 mg/L 6-BA+0.49 mg/L IBA+0.51 mg/L GA3;以1/2 MS+1.50 mg/L IBA+0.50 g/L Ac+暗培养5 d生根效果最佳,生根率达88.89%.琐琐葡萄茎段快繁的体系为:初代培养采用 MS培养基,增殖培养适宜激素浓度为6-BA 1.77 mg/L、IBA 0.49 mg/L、GA3 0.51 mg/L,生根培养适宜条件为IBA 1.50 mg/L、Ac 0.50 g/L,结合5 d暗培养处理.

This study aims to screen the optimal culture media and plant growth regulators(PGRs)for primary culture,proliferation culture and rooting culture of Vitis vinifera Suosuo,a medicinal grape culti-var endemic to Xinjiang,and to establish an efficient stem segment-based rapid propagation system for this germplasm.Single-node stem segments of Vitis vinifera Suosuo were used as explants.For primary cul-ture,the optimal basal medium for axillary bud germination was screened from five media types:MS,WPM,B5,Nitsch and ER.For proliferation culture,a quadratic regression orthogonal rotation combination design was applied to investigate the effects of different combinations of 6-benzyladenine(6-BA),indole-3-butyric acid(IBA)and gibberellic acid(GA3)on bud differentiation,proliferation and regeneration.For rooting culture,1/2-strength MS was used as the basal medium,with gradients of IBA and activated carbon(Ac)concentrations and different dark culture durations set to identify the optimal rooting formula and dark treatment conditions.The optimal medium for axillary bud germination of Vitis vinifera Suosuo stem segments was MS medium supplemented with 2.00 mg/L 6-BA,0.20 mg/L IBA,and 0.20 mg/L GA3,which achieved an axillary bud germination rate of 90.00%.The quadratic regression orthogonal rotationcom-bination analysis revealed that the order of significance of PGRs affecting stem segment proliferation was 6-BA>IBA>GA3,and the optimal proliferation formula was MS medium supplemented with 1.77 mg/L 6-BA,0.49 mg/L IBA,and 0.51 mg/L GA3.The best rooting performance was obtained with 1/2-strength MS medium supplemented with 1.50 mg/L IBA and 0.50 g/L Ac,combined with 5 days of dark culture,with a rooting rate reaching 88.89%.The optimized rapid propagation system for Vitis vinifera Suosuo stem segments is established as follows:MS medium is used for primary culture;the optimal PGR concentrations for proliferation culture are 1.77 mg/L 6-BA,0.49 mg/L IBA,and 0.51 mg/L GA3;the optimal conditions for rooting culture are 1.50 mg/L IBA,0.50 g/L Ac,combined with 5 days of dark cul-ture.
