基于DUS测试性状与ISSR标记的菊芋种质资源综合评价OA
Integrated Evaluation of Helianthus tuberosus L.Germplasm Using DUS Test Traits and ISSR Molecular Markers
为解析菊芋种质资源遗传多样性特征、挖掘优异等位基因及筛选育种材料,基于 DUS测试性状与ISSR分子标记,对40份有性繁殖和7份无性繁殖菊芋资源系统开展表型性状分析、遗传多样性分析及关联分析,以期为菊芋分子标记辅助育种提供理论依据.结果表明:5个表型性状的变异系数在15.18%~138.36%之间,单株块茎体积与单株块茎质量呈极显著正相关(P<0.01),在欧氏距离0.735 6左右处,可将菊芋品种分为 A、B两大类,在单株块茎体积和单株块茎质量两个表型性状上存在明显差异.20条ISSR引物共扩增出179条带,其中178条呈多态性,多态性比率(PPB)达99%.Popgene 32软件分析结果表明,平均等位基因数为1.990 0、平均有效等位基因数为1.539 4,平均 Nei's基因多样性指数为0.322 8,平均Shannon's信息指数为0.490 6,资源间遗传相似系数在0.514 0~0.910 6,平均为0.682 1.UPGMA聚类与群体结构分析将材料划分为3个亚群(K=3),遗传分化显著,为关联分析提供结构框架.基于 GLM 模型,共检测到62个与表型性状极显著关联(P<0.01)的位点,变异解释率为12.78%~57.80%;基于 MLM 模型,共检测到7个与表型性状极显著关联(P<0.01)的位点,变异解释率为13.46%~21.14%.综合表型和分子标记分析,筛选出高产型材料2份(青芋8号、2024JA-S23),饲用型材料3份(2024JA-S05-1、2024JA-S07、2024JA-S08),生态型材料1份(2024-2).本研究为菊芋分子设计育种与多用途产业化利用提供理论支撑和核心材料.
To elucidate the genetic diversity characteristics of Helianthus tuberosus L.germplasm re-sources,identify superior alleles,and screen breeding materials,this study conducted a comprehen-sive analysis based on DUS test traits and inter-simple sequence repeat(ISSR)molecular markers.A total of 40 sexually propagated and 7 asexually propagated germplasm accessions were systematically evaluated for phenotypic traits,genetic diversity,and marker-trait associations,with the aim of pro-viding a theoretical foundation for marker-assisted breeding of Helianthus tuberosus L.The results showed that the coefficients of variation for five phenotypic traits ranged from 15.18%to 138.36%.Tuber volume per plant and tuber mass per plant exhibited a highly significant positive correlation(P<0.01).Cluster analysis based on Euclidean distance(approximately 0.735 6)grouped the acces-sions into two major categories(A and B),which differed markedly in tuber volume and mass per plant.Using 20 ISSR primers,179 bands were amplified,of which 178 were polymorphic,resulting in a polymo-rphic percentage(PPB)of 99%.Analysis with Popgene 32 revealed an average number of al-leles of 1.990 0,an average effective number of alleles of 1.539 4,and an average Nei's gene diversity index of 0.322 8,and an average Shannon's infor-mation index of 0.490 6.The genetic similarity coef-ficient among accessions ranged from 0.514 0 to 0.910 6,with an average of 0.682 1.UPGMA cluste-ring and population structure analysis divided the germplasm into three subgroups(K=3),demon-strating significant genetic differentiation and providing a structural framework for association analy-sis.Based on the GLM model,62 loci were detected to be highly significantly associated(P<0.01)with phenotypic traits,with phenotypic variation explained ranging from 12.78%to 57.80%.Using the MLM model,7 loci were identified(P<0.01),explaining 13.46%to 21.14%of the variation.By integrating phenotypic and molecular marker analyses,two high-yielding accessions(Qingyu No.8 and 2024JA-S23),three forage-type accessions(2024JA-S05-1,2024JA-S07,and 2024JA-S08),and one ecolo-gical-type accession(2024-2)were selected.These results provide both theoretical support and core germplasm resources for molecular breeding and multi-purpose industrial utilization of Heli-anthus tuberosus L.
