Optimization of liquid bandage matrix formulation by Box-Behnken response surface methodology
目的 使用Box-Behnken响应面法优化伤筋止痛液体绷带处方.方法 以硼酸-氢氧化钠缓冲液pH、丙二醇用量、聚乙烯醇(polyvinyl alcohol,PVA)浓度、水溶性淀粉浓度为关键考察因素,构建包含黏附性能、保湿特性与交联速率3项指标的综合评价体系,其中黏附性40分,保湿特性及交联速率各30分,综合评价总分100分.采用Box-Behnken响应面法进行实验设计,通过多元回归分析优化处方参数.结果 确定最佳处方为缓冲液pH 8.9、丙二醇0.5 mL、PVA浓度20%(w/v)、水溶性淀粉15%(w/v),在此条件下综合评分为89.74分.结论 所制得的液体绷带黏性适中、保湿性良好、交联迅速.
Objective To optimize the formulation of a liquid bandage for muscular fasciitis relief using the Box-Behnken response surface methodology.Methods Key factors including the pH of the boric acid-sodium hydroxide buffer solution,the amount of propylene glycol,the concentration of polyvinyl alcohol(PVA),and the concentration of water-soluble starch were investigated.A comprehensive evaluation system was constructed based on three indicators:adhesion performance,moisturizing properties,and crosslinking rate.Among them,adhesion was worth 40 points,moisturizing properties and crosslinking rate each 30 points,and the total score of comprehensive evaluation was 100 points.The Box-Behnken response surface methodology was used for experimental design,and multivariate regression analysis was conducted to optimize formulation parameters.Results The optimal formulation was determined as a buffer solution with pH 8.9,0.5 mL of propylene glycol,20%(w/v)PVA concentration,and 15%(w/v)water-soluble starch concentration.Under these conditions,the comprehensive score was 89.74 points.Conclusion The prepared liquid bandage exhibited moderate adhesion,good moisturizing properties,and rapid crosslinking.
