益气活血中药组分对慢性脑缺血小鼠认知功能及β-淀粉样蛋白与受体RAGE/LRP-1表达的影响OA
Effects of Qi-Replenishing and Blood-Activating Traditional Chinese Medicine Components on Cognitive Function and Expression of Aβ Protein and RAGE/LRP-1 in Chronic Cerebral Ischemia Model Mice
目的 观察益气活血中药组分(人参总皂苷、银杏总酮酯和西红花总苷配伍)对慢性脑缺血模型小鼠的学习记忆能力、脑组织病理、β-淀粉样蛋白(amyloid β-protein,Aβ)及相关受体RAGE/LRP-1蛋白表达的影响.方法 将64只SPF级C57BL/6J雄性小鼠,按照随机数字表法分为假手术组、模型组、阿司匹林组和中药组分组,每组16只.采用温控电流刺激C57BL/6J小鼠双侧颈总动脉制备慢性脑缺血模型;造模成功后,假手术组和模型组采用等体积蒸馏水灌胃,阿司匹林组采用10 mg/(kg·d)阿司匹林灌胃,中药组分组采用33 mg/(kg·d)中药灌胃.给药8周后,采用Morris水迷宫方法检测学习记忆能力;HE染色观察脑组织病理形态学改变;刚果红染色观察海马CA1区淀粉样斑块沉积;蛋白免疫印迹法(Western blot)检测淀粉样蛋白1-42(amyloid β-protein 1-42,Aβ1-42)、晚期糖基化终产物受体(the receptor of advanced glycation endproducts,RAGE)、低密度脂蛋白相关受体1(low-density lipoprotein receptor-related protein 1,LRP-1)的蛋白表达水平;实时荧光定量PCR(RT-qPCR)检测Aβ1-42、RAGE、LRP-1的mRNA表达情况.结果 与假手术组比较,模型组小鼠穿越原平台次数、原平台象限游泳时间和路程均明显减少,脑组织内Aβ1-42、RAGE蛋白及mRNA表达均显著升高,LRP-1蛋白及mRNA表达降低,差异有统计学意义(P<0.05,P<0.01);HE染色可见海马CA1区神经元数量减少、胞体缩小、核膜界限不清,出现少量染色不均的淀粉样斑块沉积.与模型组比较,阿司匹林组和中药组分组小鼠穿越原平台次数、原平台象限游泳时间和路程均明显增加,Aβ1-42、RAGE蛋白及mRNA表达降低,LRP-1蛋白及mRNA表达升高,差异有统计学意义(P<0.05,P<0.01),海马CA1区神经元数量增多,细胞排列有所恢复,细胞结构稍清晰,核膜能分辨,且中药组分组上述脑组织病理改变更明显.结论 益气活血中药组分可以改善模型小鼠脑组织病理变化,调节LRP-1和RAGE的表达,减少Aβ1-42的异常沉积,改善慢性脑缺血所致的认知功能障碍.
Objective To observe the effects of qi-replenishing and blood-activating traditional Chinese medicine(TCM)components(including total ginsenosides,total Ginkgo ketone ester,and total Crocus sativus glucoside)on learning and memory ability,brain histopathology,alongside amyloidβ-protein and related receptor RAGE/LRP-1 expression in chronic cerebral ischemia model mice.Methods Sixty-four SPF grade C57BL/6J mice were divided into a sham operation group,a model group,an aspirin group,and a TCM group using the random number table method,with 16 mice in each group.A chronic cerebral ischemia model was established by stimulating the bilateral common carotid arteries of C57BL/6J mice using temperature-controlled current.After successful modelling,the sham operation group and the model group were administered distilled water of the same volume as body weight by gavage.The aspirin group was administered aspirin 10 mg/(kg·d)by gavage.The TCM group was administered the TCM components 33 mg/(kg·d)by gavage.After 8 weeks of medication,learning and memory were assessed by the Morris water maze.Brain tissue pathological changes were observed via hematoxylin-eosin(HE)staining.Amyloid plaque deposition in the hippocampal CA1 region was observed via Congo red staining.Amyloid protein(Aβ1-42),the receptor of advanced glycation end products(RAGE),and low-density lipoprotein-related protein 1(LRP-1)expression were detected by Western blot,while relevant mRNA expression was detected by real-time quantitative PCR(RT-qPCR).Results Compared with the sham-operated group,the model group significantly reduced original platform crossing numbers,along with shorter swimming time and distance traveled in the original platform quadrant.Furthermore,the model group showed significantly upregulated mRNA and protein expression levels of Aβ1-42 and RAGE in brain tissues,alongside downregulated LRP-1 protein and mRNA expression(P<0.05,P<0.01).Hematoxylin-eosin staining revealed a decreased neuron quantity in the hippocampal CA1 region,cell shrinkage,indistinct nuclear membrane boundaries,and a small amount of unevenly stained amyloid plaque deposition.In comparison to the model group,these changes,except for escape latency,were significantly reversed in both the aspirin group and the TCM group,while these brain tissue pathological changes were more significant in the TCM group(P<0.05,P<0.01).Conclusion The qi-replenishing and blood-activating TCM components can improve brain tissue pathological changes in the mice model,regulate LRP-1 and RAGE expression,and reduce abnormal Aβ1-42 deposition,thereby improving the cognitive impairment caused by chronic cerebral ischemia.
