目的:探讨针刺对乳腺癌(BC)小鼠化疗后免疫功能的调节及潜在机制.方法:将40只雌性BALB/c小鼠随机分为正常组、模型组、化疗组、针刺组、化疗+针刺组,每组8只,采用4T1-Luc细胞制备BC模型.造模成功后,化疗组予腹腔注射表柔比星,针刺组取双侧足三里、三阴交、内关、关元、气海、中脘,每天针刺30 min,化疗+针刺组给予化疗组、针刺组共同干预方式.连续干预14 d后,观察测量小鼠肿瘤体积、肿瘤质量、脾脏大小,采用ELISA测量干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)、白细胞介素-17(IL-17)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)的表达,采用流式细胞术测量小鼠脾脏CD4+T、CD8+T、Th1、Th2、Th17、Treg的细胞占比.结果:在肿瘤体积、肿瘤质量、脾脏指数方面,针刺组、化疗组、化疗+针刺组均下降,尤其化疗+针刺组下降明显.在细胞因子方面,与化疗组比较,化疗+针刺组抗癌因子IFN-γ含量升高,促癌因子IL-17、IL-10的含量降低(P<0.01),TNF-α含量升高(P<0.05).在T淋巴细胞方面,与化疗组比较,化疗+针刺组可上调CD4+T、CD8+T的占比(P<0.01),下调Th17、Treg的占比(P<0.05);与模型组比较,化疗+针刺组可上调Th1/Th2(P<0.05).结论:针刺改善BC小鼠化疗后免疫功能降低可能与调节细胞因子的表达水平,上调CD4+T、CD8+T占比,调控Th1/Th2动态平衡以及下调Th17、Treg的占比有关.

Objective:To investigate the regulatory effect of acupuncture on immune function and its underlying mechanism in mice with breast cancer(BC)after chemotherapy.Methods:Forty female BALB/c mice were randomly divided into a normal group,a model group,a chemotherapy group,an acupuncture group,and a chemotherapy+ac-upuncture group,with 8 mice in each group.A BC model was established using 4T1-Luc cells.After successful mod-eling,the chemotherapy group was intraperitoneally injected with epirubicin.For the acupuncture group,bilateral Zu-sanli(ST36),Sanyinjiao(SP6),Neiguan(PC6),Guanyuan(CV4),Qihai(CV6)and Zhongwan(CV12)were select-ed as acupoints,and acupuncture was administered for 30 minutes per day.The chemotherapy+acupuncture group received the combined interventions of the chemotherapy group and the acupuncture group.After 14 consecutive days of intervention,the tumor volume,tumor weight and spleen size of the mice were measured and observed.The ex-pression levels of interferon-γ(IFN-γ),interleukin-4(IL-4),interleukin-17(IL-17),interleukin-10(IL-10)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA).The proportions of CD4+T,CD8+T,Th1,Th2,Th17 and Treg cells in the mouse spleen were determined by flow cytometry.Results:The tumor volume,tumor weight and spleen index were all decreased in the acupuncture group,chemotherapy group and chemotherapy+acupuncture group,with the most significant reduction in the chemotherapy+acupuncture group.In terms of cytokines,compared with the chemotherapy group,the chemotherapy+acupuncture group had a significantly increased content of the anti-tumor factor IFN-γ(P<0.01),a markedly decreased content of the pro-tumor factors IL-17 and IL-10(P<0.01),and an elevated content of TNF-α(P<0.05).Regarding T lymphocytes,compared with the chemotherapy group,the chemotherapy+acupuncture group up-regulated the proportions of CD4+T and CD8+T cells(P<0.01)and down-regulated the proportions of Th17 and Treg cells(P<0.05);com-pared with the model group,the chemotherapy+acupuncture group increased the Th1/Th2 ratio(P<0.05).Con-clusion:Acupuncture ameliorates the chemotherapy-induced immune dysfunction in BC mice,which may be associated with regulating the expression levels of cytokines,up-regulating the proportions of CD4+T and CD8+T cells,modu-lating the dynamic balance of Th1/Th2,and down-regulating the proportions of Th17 and Treg cells.
